噻虫嗪对西方蜜蜂CYP6AS基因表达的影响

【摘要】 [目的]细胞色素P450单氧酶是昆虫体内重要的解毒酶。细胞色素P450 6AS亚家族基因特异性的存在于膜翅目昆虫中,一些特异性的CYP6AS基因已经被证实参与到蜜蜂对某些外源物质和杀虫剂的解毒和代谢过程中。本研究主要探测了亚致死浓度噻虫嗪对西方蜜蜂CYP单氧酶活性及CYP6AS亚家族基因表达水平的影响。[方法]首先测定了噻虫嗪LC10和LC5处理西方蜜蜂(Apis mellifera L.)后48h CYP单氧酶活性,然后应用荧光定量PCR技术检测了CYP6AS基因(CYP6AS3,CYP6AS4,CYP6AS5,CYP6AS10,CYP6AS14和CYP6AS15)表达变化情况。[结果]结果表明,噻虫嗪LC10和LC5处理后,西方蜜蜂细胞色素P450单氧酶活性均极显著增加(P<0.01)。噻虫嗪LC10和LC5处理对西方蜜蜂CYP6AS4,CYP6AS10和CYP6AS15基因表达没有显著影响;LC10和LC5处理后CYP6AS3基因表达量显著增加(P<0.05);LC10处理能够显著诱导CYP6AS5基因的上调表达(P<0.05);而LC10和LC5处理极显著抑制CYP6AS14基因的表达(P<0.01)。[结论]结果表明,噻虫嗪可能能够通过诱导CYP6AS3和CYP6AS5基因的上调表达从而增强了西方蜜蜂细胞色素P450单氧酶的活性,这为进一步探究西方蜜蜂对噻虫嗪的解毒机制提供了新的线索。
【Abstract】 [Aim] Cytochrome P450 monooxygenase is an important detoxifying enzyme in insect.The CYP6 AS subfamily are unique to hymenopterans,and several CYP6 AS genes have been implicated in detoxification and metabolism of xenobiotics and insecticides in honey bees.This study aims to investigate sublethal effects of thiamethoxam on cytochrome P450 monooxygenase activity,and expression of cytochrome P6 AS genes,in honey bees(Apis mellifera L.).[Methods]We firstly tested the activity of CYP monooxygenase in honey bees treated with LC10 and LC25 of thiamethoxam at 48 h.Then the expression levels of six CYP6 AS genes(CYP6 AS3,CYP6 AS4,CYP6 AS5,CYP6 AS10,CYP6 AS14 and CYP6 AS15)were measured by RT-qPCR.[Results] The activity of CYP monooxygenase was significantly increased with sublethal concentrations(LC10 and LC5) of thiamethoxam(P<0.01).There was no significant difference in CYP6 AS4,CYP6 AS10 and CYP6 AS15 expression compared with the control.However,the expression level of CYP6 AS3 was significantly increased by sublethal concentrations of thiamethoxam(P<0.05),and the expression level of CYP6 AS5 was induced by LC10 of thiamethoxam(P<0.05).While,the expression level of CYP6 AS14 was distinctly inhibited by LC10 and LC5 of thiamethoxam(P<0.01).[Conclusion] Our findings show that sublethal concentrations of thiamethoxam might enhance the activity of CYP by up-regulating the expression of CYP6 AS3 and CYP6 AS5,and the conclusions provide important information for exploring the detoxication mechanisms of A.mellifera L.to thiamethoxam.
