工蜂触角中哺育和采集行为相关基因的鉴定

【摘要】 蜜蜂的社会性活动主要通过嗅觉交流完成,借助不同的昆虫信息素和气味分子来实现。虽然劳动分工及主要的嗅觉器官(触角)已经被广泛研究,但是嗅觉如何影响行为的发育仍然不明确。我们对西方蜜蜂刚出房工蜂、哺育蜂及采集蜂的触角中基因的表达量进行了全面的定量分析,同时,组建了实验蜂群来采集相同日龄(10日龄和21日龄)的蜜蜂样本。RNA测序与荧光定量PCR的结果表明有6个基因与哺育行为相关,14个基因与采集行为相关,这些结果可以帮助解释嗅觉与行为变化之间的关系。这些数据可以为劳动分工中潜在的分子机制的研究提供新的观点。
【Abstract】 Honey bees,as a social insect,mainly based on olfactory communication using different pheromones and odorants.Although the division of labor and the major olfactory organ(antennae) have been extensively studied,how the olfactory influences behavioral development is still unclear.In this study,we conducted a comprehensive and quantitative analysis of gene expression in Apis mellifera antenna of newly emerged workers,nurses and foragers using transcriptome analysis.Meanwhile,we constructed experimental colonies to collect age-matched samples,including 10-day-old foragers and 21-day-old nurses,which were used to determine whether task or age is the principal determinant of differential expression.RNA sequencing and quantitative real-time polymerase chain reaction revealed that 6 and 14 genes were closely associated with nurse and forager behaviors,respectively,which could contribute to elucidating the relationship between olfactory and behavior-related changes.The data obtained in this study will provide a new perspective into the molecular mechanism underlying division of labor.
