Improvement of pulmonary function by inhaled rosuvastatin involves reversed central CD73 downregulation in asthmatic rats

【摘要】 Purpose: to test whether rosuvastatin is able to attenuate the downregulation of central CD73 and,subsequently, improve pulmonary function via attenuation of the increase of airway vagal activity.Methods: experimental rat model of allergic asthma was prepared using ovalbumin with or without simultaneous treatment with inhalation of rosuvastatin, and the effects of inhaled rosuvastatin were examined by plethysmographic measurement of pulmonary function, hematoxylin and eosin staining of the lungs, and molecular biological assay of the expression and activity of medullary CD73.Results: in ovalbumin-sensitized rats, the expression and activity of medullary CD73 were significantly decreased; ATP concentration in cerebral spinal fluid was significantly increased, and all of which were reversed in rats treated with inhaled rosuvastatin.Inhaled rosuvastatin abolished the increase of airway resistance and decrease of dynamic compliance of the lungs.Conclusion: in rat model of allergic asthma, inhaled rosuvastatin reverses the decreases in the expression and activity of brainstem CD73, and via which abolishes the increase of airway vagal activity and improves pulmonary function.
