Protein satiety factor FIT plays an essential role in feeding control and healthy ageing

【摘要】 Protein nutrient is a vital factor for animal health, while the roles of protein nutrient sensing pathway remain poorly understood.Our previous study identified a protein-specific satiety factor FIT, which suppresses protein intake upon a high level of protein consumption.In this study, we estimate the impact of this unique nutrient sensing pathway on animal health and lifespan.Compared to wild type(WT), fit mutant is more sensitive to high-protein(HP) diet.When protein nutrient content increased to two folds, the lifespan of fit mutant flies reduced significantly, accompanying with physiological and metabolic changes.In contrast, WT flies were rarely affected in this protein content.Behavioral studies showed that compared to WT flies, fit mutant flies consumed more HP food, and moreover, switched their feeding preference from HP food to normal food(NF)much slower in presence of these two optional foods.Strikingly, WT flies in 1:7 NF:HP food survived as well as those in NF, while fit mutant flies in the optional foods survived as bad as those in HP food.Thus, we suggest that defects in protein nutrient sensing lead to changes in feeding preference, and such changes in the time scale of lifetime affect the animal health and survival at a significant level.
