金黄仓鼠高血脂模型建立及相关指标检测

【摘要】 目的建立金黄仓鼠高血脂动物模型,为进一步研究血脂代谢机制奠定基础。方法将金黄仓鼠分为普通饮食对照组和高脂饮食模型组,分别用普通饮食和高脂饮食饲养12周,于实验4周、8周和12周称量动物体质量和检测血清甘油三酯(TG)、血清总胆固醇(TC)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)水平。实验结束对肝脏进行形态学观察,应用荧光定量PCR技术检测载脂蛋白A5(Apoa 5)、过氧化物酶增殖激活的受体α(PPARα)在肝脏表达情况。结果在本研究结果显示,模型组体质量明显高于对照组(P<0.05);模型组TG、TC、LDL-C、HDL-C显著升高(P<0.05),肝脏出现脂肪肝样改变,通过荧光定量PCR分析了Apoa5、PPARαmRNA在肝脏组织的基因相对表达变化。模型组肝脏Apoa5、PPARαmRNA与对照组比较表达量有明显下调趋势,与TG水平呈负相关。说明Apoa5、PPARα与血脂代谢异常有密切联系,TG水平升高可能会引起Apoa5、PPARα水平降低,从而导致血脂代谢异常,反之,血脂代谢异常会引起ApoA5水平降低,从而导致TG水平升高,三者间互为因果,共同导致血脂异常的发生发展。有研究发现他汀类药物降TG的作用是通过上调PPARα进而升高Apoa5的表达水平达到降低TG。模型组肝脏Apoa5、PPARαmRNA与对照组比较表达量明显降低。结论成功建立黄金仓鼠高血脂动物模型,Apoa5、PPARα可能是血脂调节的关键因子。
【Abstract】 Objective To establish an animal model of hyperlipidemia in golden hamster,in order to la y a foundation for further study on the mechanism of blood lipid metabolism.Methods:The hamst ers were divided into normal diet group and high-fat diet group,which were fed with normal diet a nd high-fat diet for 12 weeks respectively.Their body weights and level of blood lipids,including trig lycerde(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C) and High Density Lip oprotein Cholesterol(HDL-C),were detected on the 4,8 and 12 weeks after experiment.At the end of the experiment,the morphology of the livers were observed;the expression of apolipoprotein A5(Apoa5) and peroxisome proliferator activated receptors α(PPARα) in livers were detected by fluor escence quantitative PCR.Results:Body weights in high-fat diet group were significantly higher tha n those in the control group(P < 0.05).The levels of TG,TC,LDL-C and HDL-C in high-fat diet group w ere significantly increased than those in the control group(P < 0.05).The livers showed fatty liver lik e changes in the high-fat diet group.The mRNA expression of Apoa5 and PPARα were significantly d ecreased than those in the control group in liver.Conclusion:The animal model of hyperlipidemia i n golden hamster was successfully established.Apoa5 and PPARα may be the key factors of blood li pid regulation.
