帕金森病异动症模型大鼠纹状体内多巴胺D1、D2受体变化及复方地黄方的干预作用

【摘要】 目的:研究复方地黄方对阴虚动风证帕金森病异动症大鼠多巴胺D1、D2受体表达的影响。方法:采用经典的6-OHDA脑立体定向注射术建立大鼠PD模型,在此基础上腹腔注射2周左旋多巴/苄丝肼制备异动症模型。把成功的异动症模型模型分为LID模型组4W、6W组和复方地黄方组4W、6W组,同时纳入正常对照组4W、6W组和假手术组4W、6W组,每组6只。在4W、6W不同时间点,运用Western Blot方法测定各组大鼠纹状体多巴胺D1、D2受体的表达。结果:LID大鼠纹状体中D1、D2受体的含量均增加。与LID组比较,复方地黄方4周、6周组D1、D2受体的表达均有下降趋势,差异有统计学意义(P<0.001);复方地黄方6周组与4周组相比较,D1受体的表达有下降趋势,差异有统计学意义(P<0.001),D2受体的表达有下降趋势,无统计学意义。结论:LID大鼠直接通路与间接通路活动均增强,以直接通路活动增强为主,复方地黄方通过对D1、D2受体的调节,对直接通路和间接通路的失衡有一定的改善作用。

【Abstract】 Objective To approach the effect of compound rehmannia prescription on the expression of dopamine D1 and D2 Receptors of Dyskinesia Model Rat with Parkinson’s Disease. Methods PD model rats were made by the classic injection of 6–hydroxy dopamine to destroy the substantia nigra with brain stereotaxic instrument. Then PD model rats were made LID model rats with Parkinson’s disease through the intraperitoneal injection of LD & benserazide for 2 weeks. The successful LID models were randomized into LID model for 4 and 6 weeks and compound rehmannia prescription groups for 4 and 6 weeks,and normal control,sham-operated groups for 4 and 6 weeks were brought into,6 rats in each group. The expression of dopamine D1 and D2 receptors in the rats’ destroyed lateral striatum was examined by means of Western Blot. Results In LID model group, the expression of dopamine D1 receptor showed an increasing trend compared with that in the normal control group.,As the time passed after the model establishment, the expression of dopamine D1 receptor increased gradually. The expression of dopamine D2 receptor showed an increasing trend compared with that in the normal control group. As the time passed after the model establishment, the expression of dopamine D2 receptor decreased. Conclusion Direct pathway and the indirect pathway activities of LID rats are reinforced. Dopamine D1, D2 receptor abnormalities are a key link in its incidence. Compound rehmannia prescription balanced changes in dopamine D1 and D2 receptors to alleviate Parkinson’s disease dyskinesia.
