Mongersen对大鼠牙周炎的抗炎效果观察

【摘要】 目的:在体内观察mongersen对大鼠牙周炎的抗炎效果。方法:选取健康8周龄雄性Wistar大鼠22只,从中随机选取16只,用正畸丝结扎于其上颌双侧第一磨牙牙颈部构建大鼠牙周炎模型,剩余6只为空白对照(C0组),6周后从牙周炎模型组与健康组各随机选取1只处死并验证大鼠牙周炎模型已成功建立。随后拆除结扎丝,将15只牙周炎大鼠随机分3组,每组5只,其中一组在上颌双侧第一磨牙颊、腭侧牙龈各注射25ng/μl的mongersen 20μl,一周3次(M组);另一组在相同部位注射等量生理盐水(N组);剩余5只为牙周炎对照组,不做任何处理(C组)。注射药物2周检测各组大鼠上颌第一磨牙牙周附着丧失、牙周探诊深度、探诊出血等牙周炎相关临床指标后将其处死,qRT-PCR检测各组大鼠牙龈组织炎症因子基因的表达情况。结果:结扎6周后,大鼠牙龈红肿,牙周袋形成,且X-ray检测发现有牙槽骨吸收,即成功构建大鼠牙周炎模型。将其分组并注射药物2周后,牙周临床指标显示mongersen组(M组)相比牙周炎对照组(C组)及生理盐水组(N组)牙周探诊深度、附着丧失及探诊出血症状减轻,且肉眼见M组相比C组及N组牙龈组织炎症缓解,探诊牙龈质地坚韧;qRTPCR结果显示盐水组(N组)及牙周炎组(C组)与空白对照组(C0组)比TNF-α、IL-1β、IL-6等炎症因子基因的表达明显升高,而加入mongersen组(M组)与盐水组(N组)及牙周炎组(C组)相比TNF-α、IL-1β、IL-6等炎症因子基因的表达显著降低,结果均有明显差异(P<0.001)。结论:mongersen可明显缓解大鼠牙周炎模型中牙周组织炎症程度且能显著降低其牙周组织内炎症因子基因的表达。

【Abstract】 Objective:To observe the anti-inflammatory effect of mongersen on periodontitis rats in vivo.Methods:Twenty-two healthy male Wistar rats aged 8 weeks were randomly selected.Then randomly selected 16 rats from them,ligature orthodontic silk to the neck of the maxillary first molar to construct the model of periodontitis,and the remaining 6 rats were negative controls(Group C0).Six weeks later,one rat was respectively selected from the periodontitis model group and the blank group to confirm the model of periodontitis had been established.Then removed the orthodontic silk and randomly divided 15 periodontitis rats into 3 groups,5 rats in each group,injected mongersen(25 ng/μl;20 μl/time;Group M) and the saline into the buccal and lingual gum of maxillary first molars,3 times a week,2 weeks in total.And the remaining 5 rats were periodontitis control group(Group C).And then detected the periodontal attachment loss,periodontal probe depth,probe bleeding index of the maxillary first molar in the periodontitis rats,and then all of the rats were sacrificed,the expression of inflammatory cytokines in gingival tissues of rats in each group were detected by qRT-PCR.Results:After 6 weeks of ligation,the experimental rats ’ gingival tissue was inflamed and swollen,simultaneously they formed periodontal pockets and alveolar bone resorption.That is,the rat periodontitis model was successfully constructed.Two weeks after the injection of drugs,clinical indicators showed that in mongersen group(Group M) the bleeding symptoms,the depth of the periodontal probe and the attachment loss were alleviated,at the same time visual observation of the Group M had reduced inflammation of the gingival tissue compared with the periodontitis control group(Group C) and the saline group(Group N),and their texture of the gingival arounding the maxillary first molar was tough.The qRT-PCR results showed that the gene expression of the inflammatory factors such as TNF-α,IL-1β and IL-6 in the saline group and periodontitis control group were significantly increased,compared with the blank control group;and the mongersen group significantly decreased the gene expression of inflammatory factors compared with the saline group and periodontitis control group(P<0.001).Conclusions:Mongersen can significantly alleviate periodontal inflammation in periodontitis rats and can significantly reduce the gene expression of inflammatory factor in periodontal tissues.
