【摘要】 目的:黄芩有效部位可促进牙周膜细胞的增殖能力和成骨活性作用,现为观察其在整体水平的治疗效果,将其制备成缓释凝胶并用于小型猪实验性牙周炎的局部治疗。方法:将黄芩有效部位制备成缓释凝胶,并优化处方浓度、检测体外释放、流变学和微观学观察;将5只小型猪4个区的P1、P2、M1和M2根分叉去骨、牙颈部结扎制备实验性牙周炎模型,8周后待建模成功,采用不同分组(空白凝胶组、药物凝胶组、阳性对照组)局部治疗,12周后检查牙周各项指数、ELISA分别检测IL-8和PGE2含量、分离颌骨组织行HE染色、免疫组化染色分析等。结果:制备出性状良好2%黄芩有效部位凝胶,体外释放能力、流变学指标、微观结构良好;实验性牙周炎建模成功后进行局部治疗,检测牙周各项指数、细胞因子、HE染色和免疫组织化学染色等都说明黄芩有效部位凝胶对牙周炎的抗炎和成骨有一定的促进作用。结论:2%黄芩有效部位原位凝胶对牙周炎抗炎和成骨有促进作用,可以作为一种新的治疗牙周炎的药物用于临床。更多还原

【Abstract】 OBJECTIVES:The effective parts of Scutellariabaicalensis can promote the proliferation and osteogenic activity of periodontal ligament cells in vitro.In order to observe the therapeutic effect of the living standard,the gel was prepared for the topical treatment of experimental periodontitis in miniature pigs.METHODS:Preparation of the effective parts of Scutellariabaicalensis into a sustained release gel,then optimize prescription concentrations,detect the release in vitro,rheology index and microscopic observations.P1,P2,M1 and M2 teeth in4 districts of 5 miniature pigs were prepared for the experimental periodontitis model by boneless in root bifurcated and ligated in the neck of teeth.After 8 weeks,the models were built successfully,and then local treated with different groups(blank gel group,medicine gel group,positive control group).12 weeks later,the pigs were conducted with periodontal examination,detection of IL-8 and PGE2 content by ELISA,and isolated the jaw bone tissues for HE staining,immunohistochemical staining analysis and so on.RESULTS:The 2% effective parts of Scutellariabaicalensis gel was prepared,the release capacity in vitro,rheological parameters,microstructure were fine.Local treatment after the experimental periodontitis modeling successfully prepared,the 2% effects of Scutellariabaicalensis gel was effective in the treatment of periodontitis through detecting the periodontal indexs,cell factors,HE and histochemical staining.CONCLUSIONS:2% effective parts of Scutellariabaicalensis gel has promotion effect to the inflammation and bone formation of periodontitis,so it could be used as a new drug for the treatment of periodontitis.更多还原
