【摘要】 目的:探讨TOLL样受体4(Toll-like receptor 4,TLR4)基因的单核苷酸多态性(Single Nucleotide Polymorphisms,SNPs)与慢性牙周炎的关系。方法:选取241例来自北京石景山社区中定期体检的队列患者,采集颊拭子,收集非刺激性的静息全唾液,再进行全口牙周检查记录牙周临床指标。通过Sequenom公司的MassARRAY进行颊拭子DNA的TLR4 SNPs检测,包括rs11536889,rs1927906,rs1927911,rs2149356,rs4986790,rs4986791,rs6478317,rs7873784,分析其等位基因和基因型频率的分布。通过聚合酶链式反应(polymerase chain reaction,PCR)检测唾液沉淀中的Pg含量。分析TLR4的SNPs与慢性牙周炎程度及其Pg感染的关系。结果:241名患者的8个TLR4的SNPs分布总体并没有显著的统计学差异。而对于有Pg感染的患者,3个SNPs位点rs1927911 (TT/CC+CT),rs2149356 (AA/CC+CA)和rs6478317 (GG/AA+AG),在中重度牙周炎患者中分布明显增高,较非牙周炎或轻度牙周炎患者有显著的统计学差异。结论:在中国人群,TLR4的3个SNPs(rs1927911,rs2149356,rs6478317)可增强Pg感染的中重度慢性牙周炎的易感性。更多还原

【Abstract】 Objective:To investigate the correlation between the Single Nucleotide Polymorphisms(SNPs) in toll-like receptor 4(TLR4) gene and the susceptibility of chronic periodontitis.Methods:241 Chinese participants from the cohort of Beijing Shijingshan Community were recruited.Buccal swab samples,the whole unstimulated saliva and periodontal clinical parameters were collected.Human DNA extracted from buccal swab samples were used for genotyping 8 SNPs of the TLR4 gene(rs11536889,rs1927906,rs1927911,rs2149356,rs4986790,rs4986791,rs6478317,rs7873784)by the Sequenom MassARRAY system.Porphyromonas gingivalis(Pg) was detected from the deposition of the whole unstimulated saliva through polymerase chain reaction(PCR) method based on 16 S r RNA.The correlation between SNPs of TLR4 and chronic periodontitis susceptibility in patients who are detected with or without Pg was investigated.Results:There was no significant difference in the distribution of the 8 SNPs of TLR4 in 241 Chinese participants.When combining with Pg infection,rs1927911(TT/CC+CT),rs2149356(AA/CC+CA),rs6478317(GG/AA+AG) were each independent risk factors.Conclusion:SNPs of TLR 4:rs1927911,rs2149356,rs6478317 were associated with moderate/severe periodontitis in Chinese population infected with Pg.更多还原
