梅花鹿茸胶梅原蛋白抗菌肽的制备及其抑菌活性测定

【摘要】 对梅花鹿茸胶原蛋白经木瓜蛋白酶水解后得到鹿茸胶原多肽,以水解度为指标,确定最佳酶解条件为:温度50℃、PH值6.0、酶解时间5h、最适酶用量为6000U/g.按此条件提取得到的梅花鹿茸胶原多肽水解度为15.49%;采用SDS-PAGE凝胶电泳的方法测定梅花鹿茸胶原多肽的分子量集中在6.5kD以下,然后对胶原多肽酶解液采用SephadexG-25柱凝胶层析技术进行分离纯化后测定所制得的小分子胶原蛋白肽对大肠杆菌及金黄色葡萄球菌具有很好的的抑菌活性,抑菌率分别达到83.33%和69.23%。
