清胰颗粒对重症急性胰腺炎大鼠STAT3表达的影响
Effects of Qingyi Granules(清胰颗粒) on the STAT3 expression in pancreatic tissue of rats with severe acute pancreatitis

【摘要】 目的:探讨清胰汤对重症急性胰腺炎(SAP)大鼠炎症反应及信号转导与转录激活因子(STAT3)蛋白表达的影响。方法:54只雄性SD大鼠随机分为3组:假手术组、SAP模型组、清胰汤组,每组18只。采用3.5%牛黄胆酸钠胰胆管注射法制备SAP大鼠模型,实验前2h分别灌胃给予生理盐水或清胰颗粒水溶液,以后每12h灌胃一次。各组大鼠造模后按不同时间,即6、12h、24h取材。取血用于测定血清肿瘤坏死因子α(TNF-α)、白介素6(IL-6)水平;取胰腺组织行HE染色检测胰腺病理改变,并用试剂盒检测胰腺组织髓过氧化物酶(MPO)活性,用Western blotting方法胰腺组织总STAT3和磷酸化STAT3蛋白(p-STAT3)的蛋白表达。结果:在12、24h时与模型组比较清胰颗粒能显著降低SAP大鼠血清TNF-α、IL-6水平,减轻胰腺炎细胞浸润及减轻肺组织病理评分,降低肺组织MPO活性,造模24h后与模型组比较清胰颗粒能明显降低肺组织STAT3,p-STAT3蛋白表达水平。结论:清胰颗粒通过降低SAP大鼠血清TNF-α、IL-6水平和肺组织MPO活性,减轻胰腺过度的炎症反应和胰腺组织损伤,这种作用保护作用可能与减少胰腺组织STAT3,p-STAT3蛋白表达有关。
【Abstract】 Objective:To investigate the effects of Qingyi Granules on the STAT3 expression in rats with severe acute pancreatitis.Methods:Fifty-four Wistar male rats were randomly divided into three groups:sham operation group,SAP model group,SAP+Qingyi granules group,(n=18).The model of SAP was induced by injecting 3.5%sodium deoxycholate into pancreatic and biliary duct,the sham operation group were injected only with saline.The Qingyi granules or saline was perfused into the stomach in SAP+Qingyi granules group or the SAP model group respectively,at 2 hour before induction of SAP.At 6,12,24 h after induction of SAP,the serum and pancreas tissues were harvested.The levels of TNF-α and IL-6 in serum were detected by ELISA method.The histopathological evaluation and the myeloperoxidase(MPO) activity were detected in pancreas tissues.The protein expression of STAT3 and D-STAT3 in pancreas tissues was determined by Western blotting.Results:Compared with SAP model group.the serum levels of TNF-α and IL-6 was decreased,the histopathological score and the MPO activity of pancreas tissues were reduced in the SAP+Qingyi granules group at 12,24 h(P<0.05).At 24 h after induction of SAP.the protein levels of STAT3 and p-STAT3 was decreased in Qingyi granules group,compared with SAP model group(P<0.05).Conclusions:Qingyi granules can attenuate the early damage of pancreas tissues in rats with SAP via lowering the levels of serum TNF-α,IL-6 and inhibiting STAT3 signaling pathway in pancreatic tissues.
