两种食药用真菌菌丝胞内多糖提取方法的研究
Optimum Extracting Technology of Intracellular Polysaccharides from Two Edible-medicinal Fungi Mycelium

【摘要】 采用正交实验研究热水浸提法提取蛹虫草和云芝菌丝胞内多糖的最佳条件,并以此为基础研究了热水浸提法、冷热交替浸提法、盐酸浸提法、氢氧化钠浸提法对蛹虫草菌丝胞内多糖提取率的影响。结果表明,蛹虫草菌丝热水浸提的最佳提取条件为:料液比1:40 (g:mL),提取温度75℃,提取时间2.5h,提取次数2次,此条件下菌丝胞内多糖的提取率为14.74%;云芝菌丝热水浸提的最佳提取条件为:料液比1:30 (g:mL),提取温度90℃,提取时间2h,提取次数2次,此条件下菌丝胞内多糖的提取率为5.38%。按照料液比1:40 (g:mL),热冷交替浸提法(热水浸提2.5h+冷水浸提48h)对蛹虫草菌丝胞内多糖提取率最高,为15.92%。按照热水浸提的最佳提取条件,不同浓度的酸、碱对蛹虫草菌丝胞内多糖提取率的影响不同,浓度为55g/L的氢氧化钠浸提蛹虫草菌丝胞内多糖的提取率最高,为6.31%;浓度为25g/L的盐酸浸提蛹虫草菌丝胞内多糖的提取率最高,为9.99%。不同的提取方法对蛹虫草菌丝胞内多糖提取率的影响不同,冷热交替浸提法(热浸2.5h+冷浸48h)对蛹虫草菌丝胞内多糖提取效率最高,其次是热水浸提法(热浸2.5h, 2次)、盐酸浸提法(25g/L)和氢氧化钠浸提法(55g/L)。
【Abstract】 The optimized extraction conditions of intracellular polysaccharides from Cordyceps militaris and Coriolus versicolor mycelium by hot water extraction has been investigated using orthogonal array design L9(34) and optimization of extraction parameters were used to further analysis the effect of hot water extraction, thermal cycling extraction, hydrochloric acid extraction, and sodium hydroxide alkali extraction on extraction ratio of intracellular polysaccharides from C. militaris mycelium. The results showed that the optimum extraction conditions of intracellular polysaccharides by hot water extraction were established as follows: solid-liquid ratio of 1:40(g: m L), 75 ℃, 2 extraction cycles, and 2.5 h each cycle for C. militaris; and solid-liquid ratio of 1:30(g: m L), 90 ℃, 2 extraction cycles, and 2 h each cycle for C.versicolor. The corresponding extraction ratio of intracellular polysaccharides from these two edible-medicinal fungi were 14.74% and 5.38%, respectively. The optimum extraction conditions of thermal cycling extraction method consisted of hot water extraction 2.5h in optimum condition and cold water extract 48 h subsequently, of which extraction ratio of 15.92%. When the treatment concentration of sodium hydroxide and hydrochloric acid reached 55g/L and 25g/L, alkali extraction and acid extraction exhibited the highest extraction ratio of 6.31% and 9.99%, respectively.The results indicated the optimum extraction method is thermal cycling extraction,followed by hot water extraction, cold water extraction hydrochloric acid extraction and sodium hydroxide alkali extraction.
