基于微孔板培养及检测技术的高产色素低产桔霉素的红曲霉快速筛选

【摘要】 高产色素低产桔霉素的红曲霉具有重要的生产应用价值。然而,现今红曲霉的培育与筛选仍然较为繁琐。本研究建立了微孔板培养体系,以生物量、色素量、桔霉素量为指标同时跟踪了红曲霉在微孔板和摇瓶中的发酵过程,以相关系数来说明微孔板培养的可行性;通过动力学方程来描述微孔板培养体系中红曲霉生物量与色素及桔霉素产量之间的关系,并确定最佳培养周期;在此基础上用微孔板对84株红曲霉进行培养,分别采用比色法和酶联免疫法对红曲色素和桔霉素进行快速测定,获得高产色素低产桔霉素的红曲霉菌株。结果表明:微孔板与摇瓶体系的生物量、色素量、桔霉素量的变化趋势一致,相关系数均在0.8以上,两体系高度正相关;由发酵动力学方程得知,在发酵至第3天时,菌株进入生长稳定期,此时红曲色素与桔霉素快速累积,在发酵至第5天时,红曲霉的色素及桔霉素产量均达最高值并随着发酵周期的延长产物积累量保持不变,确定发酵周期为5天;根据检测结果,在84株菌中获得一株高产红曲色素低产桔霉素的红曲霉ZH,并通过形态学和分子生物学鉴定其为紫色红曲霉。微孔板培养及检测体系对于菌株筛选快速而便捷。
【Abstract】 Monascus that can produce higher-yield pigment and lower-yield citrinin is with great value.However,the cultivation and screening of Monascus are still complicated.In this study,the fermentation process of Monascus in microplates was analyzed using the indicators of biomass,pigment,and citrinin content,and compared with shake-flask fermentation to explain the feasibility of microplate culture.The dynamic equation of Monascus microplate culture was established,and used for describing the relationship between biomass and metabolites to obtain the optimal fermentation period.On this basis,84 strains of Monascus were cultivated in microplates.Contents of pigment and citrinin were determined by colorimetry and Elisa,and then screen out the best Monacus.The results showed that the change of biomass,pigment and citrinin content in microplate system strongly coincided with those of shake-flask system.According to the dynamic equation,the pigment and citrinin accumulated rapidly during the stable growth stage of Monascus when fermented at the third day.The production of pigment and citrinin of Monascus reached the maximum at the fifth days and remained afterwards,which suggested a fermentation period of 5 days.An Monascus named ZH was selected due to its higher-yield pigment and lower-yield citrinin,and then it was identified as Monascus parpureus using morphological and molecular biology methods.The microplate culture and detection system is fast and convenient for screening strains.
