乳酸菌喷雾干燥相关组学研究

【摘要】 乳酸菌喷雾干燥较现在应用广泛的冷冻干燥,有高效低耗和可连续生产的特点。但由于其过程中存活率低,并没有大量投入生产。目前对乳酸菌喷雾干燥的研究主要集中在优化干燥前发酵参数及干燥过程参数、保护剂的开发和干燥前触发乳酸菌应激反应方面。对喷雾干燥组学方面有相关研究,但研究较少,也没有系统的总结。因此本文对乳酸菌已有的转录组和全基因组相关研究进行了综述。当前转录组-表型匹配方法的研究较多,对全基因组也有相关研究。发现与胁迫相关的基因和新基因及metC-cysK操纵子可能作为筛选健壮菌株的指标,并帮助选择发酵参数。证明可能个别氨基酸代谢在氧化胁迫存活中起到关键作用。但对蛋白组学的研究甚少,未来可通过转录组与蛋白组相结合的方法进行相关研究。喷雾干燥热胁迫和氧化胁迫的组学研究较多,但对干燥应激和整个喷雾干燥过程各种胁迫共同作用研究的组学较少。

【Abstract】 Lactic acid bacteria spray drying is characterized by high efficiency,low consumption and sustainable production compared to the current widely used freeze-drying.However,due to the low survival rate in the process,it has not been put into production.At present,the research on spray drying of lactic acid bacteria mainly focuses on optimizing the fermentation parameters before drying and the parameters of drying process,the development of protective agent and the triggering of lactic acid bacteria stress reaction before drying.There were few studies on spray drying omics,and there was no systematic summary.Therefore,this paper reviewed the existing transcriptome and genome-wide studies of lactic acid bacteria.There were many studies on the current transcriptome-phenotypic matching method,and there were related studies on the whole genome.It was found that genes associated with stress and new genes and the metC-cysK operon may serve as indicators for screening robust strains and help select fermentation parameters.It was demonstrated that individual amino acid metabolism may play a key role in the survival of oxidative stress.However,there was little research on proteomics.In the future,related research can be carried out by combining transcriptome and proteome.There were many omics studies on spray drying heat stress and oxidative stress,but there were few constitutive studies on dry stress and the combined effects of various stresses in the whole spray drying process.
