芡实抗性淀粉的制备及其消化特性研究

【摘要】 芡实为一年生大型水生草本作物芡的果实,药食两用,富含淀粉,具有较大的开发潜力与市场前景。如今,用于抗性淀粉工业化生产的原料较为单一,且高纯度抗性淀粉产品较为缺乏。本研究以苏州产地芡实为原料,采用普鲁兰酶-压热法制备高纯度RS3型抗性淀粉,并优化了制备工艺。首先,通过单因素试验,研究淀粉乳浓度、普鲁兰酶作用温度、普鲁兰酶作用时间、压热温度和压热时间对抗性淀粉得率和纯度的影响。接着,通过正交实验和响应面法分析可得普鲁兰酶-压热法制备芡实抗性淀粉的最佳工艺为:淀粉乳浓度26.5%,普鲁兰酶用量4 U/g,普鲁兰酶作用时间2.3 h,压热温度120℃,压热时间23 min。在该工艺条件下制得的芡实增抗淀粉中抗性淀粉含量为(18.34±0.21)%,经纯化后其抗性淀粉纯度可达(84.00±0.52)%。体外消化模拟实验表明,经纯化后的芡实抗性淀粉在三段式模拟消化(口腔、胃、小肠)中,可一定程度降低大米淀粉与小麦淀粉的消化速率。上述制备方案为高纯度抗性淀粉的生产提供了一定的理论基础和实践经验,在控糖食品或营养保健食品中有较为广阔的应用前景。
【Abstract】 Euryale Ferox Salisb is an annual aquatic herbaceous plant,which produces starchy seeds named euryale ferox.Euryale ferox are edible with high health value,therefore,the development prospects are broad with great market potential.Nowadays,herb species used for production of commercial resistant starch(RS) are few and high-purity resistant starch products are hardly to be found.In this research,euryale ferox from Suzhou were selected as raw material for preparation of RS3-type resistant starch with the debranching-autoclaving method.The effects of starch concentration,pullulanase dosage and reactive time,autoclave temperature and time on the yield and purity of euryale ferox resistant starch were studied through single-factor tests.Based on the response surface methodology,the optimal conditions for preparation of euryale ferox resistant starch were as follows:starch concentration 26.5%,pullulanase dosage 4 U/g,pullulanase response time 2.3 h,autoclave temperature 120 ℃ and autoclave time 23 min.Following the processing parameters shown above,the content of resistant starch in resistance-enhanced euryale ferox starch was(18.34 ± 0.21)%.With additional purification processing,the purity of euryale ferox resistant starch was(84.00 ± 0.52)%.During oral-stomach-small intestine digestion in vitro,the digestive rate of rice starch and wheat starch could be reduced to some extent with the addition of purified euryale ferox resistant starch.This research could provide theoretical basis and practical experience for the production of high-purity resistant starch,which could be used in the formulas of diabetes-controlling food or nutraceuticals with spacious market prospect.
