基于MBISA修饰纳米颗粒提取溶菌酶新方法

【摘要】 本研究采用共沉淀法制备了磁性Fe3O4纳米粒子,在其表面进行Au3+还原,获得Fe3O4/Au纳米粒子,并利用2-巯基-5-苯并咪唑磺酸钠(2-Mercapto-5-benzimidazolesulfonic acid,MBISA)对Fe3O4/Au进行修饰,制备得到Fe3O4/Au-MBISA复合纳米颗粒.应用光子相关光谱(PCS)和振动样品磁强计(VSM)等对Fe3O4/Au-MBISA复合纳米颗粒进行表征,结果显示该复合纳米颗粒的平均水合粒径为74 nm,具有超顺磁性,饱和磁化强度(Ms)为44 emu/g.MBISA修饰使复合颗粒的等电点(pI)由pH=5.7变为3.6,在较大pH值范围内其表面都保持较高的负电性.研究了Fe3O4/Au-MBISA对溶菌酶的吸附及解吸特性,包括离子强度、吸附时间、温度、pH等因素的影响.结果显示Fe3O4/Au-MBISA复合纳米颗粒对溶菌酶的最大吸附量为346μg/mg.在最佳释放条件下,Fe3O4/Au-MBISA复合纳米颗粒的溶菌酶释放率可达100%.Fe3O4/Au-MBISA复合纳米颗粒具有良好的重复利用性能,经过7次吸附-洗脱循环后,其溶菌酶吸附量仍可保持在91%以上.将Fe3O4/Au-MBISA复合纳米颗粒应用于蛋清溶菌酶的提取纯化,结果显示,所提取到的溶菌酶具有较高的纯度,SDS-PAGE呈单一条带.综上所述,Fe3O4/Au-MBISA复合纳米颗粒是一种溶菌酶的良好载体,可应用于蛋清中溶菌酶的提取纯化研究及生产中。

【Abstract】 2-Mercapto-5-benzimidazolesulfonic acid(MBISA) modified Fe3O4/Au nanoparticles(Fe3O4/Au-MBISA nanoparticles) were synthesized in aqueous solution and characterized by photo correlation spectroscopy(PCS) and vibrating sample magnetometer(VSM).The average diameter of Fe3O4/Au-MBISA nanoparticles was 74 nm with a polydisperion(PDI) of 0.244,and the nanoparticles behaved superparamagnetically with saturated magnetization(Ms) of 44 emu/g.The isoelectric point(pI) of the Fe3O4/Au-MBISA nanoparticles was located at pH 3.6 and a highly negative surface charge in a wider range of pH was observed.The so-obtained Fe3O4/Au-MBISA nanoparticles were capable of specific adsorbing lysozyme.The maximum amount of lysozyme adsorbed on 1.0 mg Fe3O4/Au-MBISA nanoparticles was 346 μg.The lysozyme desorption behavior was studied and the lysozyme recovery from Fe3O4/Au-MBISA nanoparticles approached 100% under optimal conditions,and the reusability studies showed that the nanoparticles could maintain about 91% of the initial lysozyme adsorption capacity after 7 repeated adsorption-elution cycles.The Fe3O4/Au-MBISA nanoparticles were used in the purification of lysozyme from chicken egg white,which was verified by a single SDS-PAGE band.Therefore,the obtained Fe3O4/Au-MBISA nanoparticles exhibited excellent performance in the direct purification of lysozyme from egg white.
