酵母β-葡聚糖对高脂血症大鼠肠屏障功能的影响

【摘要】 在建立高脂血症大鼠模型的基础上,探讨酵母β-葡聚糖对肠屏障功能的影响。将48只正常/高脂血症SD大鼠分为对照组(C,普通日粮)、高脂血症模型组(HL,高脂日粮)、高脂血症酵母β-葡聚糖处理组(HY,添加5%酵母β-葡聚糖的高脂日粮)和酵母β-葡聚糖组(YG,添加5%酵母β-葡聚糖的普通日粮),经为期3周饲养实验后,分析生长和肠屏障功能相关指标。与C组相比,HL组大鼠体重显著降低,血清甘油三酯和总胆固醇水平显著增加(P<0.05);空肠黏膜结构破坏,肠上皮细胞凋亡率显著增加(P<0.05),二胺氧化酶含量、紧密连接蛋白(ZO-1、occludin、JAM)表达水平明显降低。与HL组相比,HY组大鼠血清中甘油三酯和胆固醇水平显著降低(P<0.05),高脂血症导致的空肠结构破坏、肠上皮细胞凋亡增加、二胺氧化酶含量及紧密连接蛋白表达水平的降低均得到显著改善(P<0.05)。日粮中添加5%的酵母β-葡聚糖能够显著缓解由高脂血症诱发的肠屏障损伤,对维持肠道健康具有积极作用。

【Abstract】 Based on the establishment of the rat model of hyperlipidemia,this study researched the effect of yeast beta-glucan on intestinal barrier function.Forty-eight Sprague Dawley rats were divided into control group(C,normal diet),hyperlipidemia model group(HL,high fat diet),hyperlipidemia rats fed with yeast β-glucan group(HY,high fat diet with 5% yeast beta-glucan) and yeast β-glucan group(YG,normal diet with 5% yeast beta-glucan).After three weeks,growth and intestinal barrier function indicators were analyzed.The results indicated that compared with the control group,the weight of the rats in the HL group decreased significantly,triglyceride and total cholesterol levels increased significantly(P < 0.05);jejuna mucosal structure were destroyed and the intestinal epithelial cell apoptosis rate increased significantly(P <0.05);diamine oxidase level and tight junction proteins(ZO-1,occludin,JAM) expression level were decreased significantly(P <0.05) in HL group rats.Compared with HL group,the triglyceride and cholesterol levels in rat serum of HY group rats were reduced significantly(P < 0.05);the destruction of jejunum morphological structure,the apoptosis of intestinal epithelial cell,and the reduction of diamine oxidase amount and tight junction protein expression level in HY group rats were improved dramatically.These results suggested that the addition of 5% yeast beta-glucan to the diet could alleviate the intestinal barrier damage induced by hyperlipidemia,and exert positive effect on the maintenance of intestinal health.
