苦瓜发酵多糖对肥胖大鼠的血脂调节作用

【摘要】 目的:利用植物乳杆菌NCU116发酵苦瓜并提取多糖,研究苦瓜发酵多糖对高脂膳食诱导的肥胖大鼠血脂调节作用及其机制。方法:设立正常组(Normal)、模型组(Model)、苦瓜发酵多糖组(FP)、苦瓜未发酵多糖组(NFP),分别灌胃生理盐水与多糖8周,剂量为10 mL/kg;并记录体重及生理状况;测定其血脂指标;评估瘦素(Leptin)和脂联素(Adiponectin)水平;并做附睾脂肪病理切片分析;探究肥胖大鼠的脂质组学并研究其脂质代谢。结果:肥胖大鼠灌胃苦瓜发酵多糖后,体重显著降低,脂肪细胞肥大症状明显减轻。总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDLC)含量显著降低,高密度脂蛋白胆固醇(HDL-C)含量升高(P<0.05);瘦素(Leptin)水平升高,脂联素(Adiponectin)水平降低;此外,苦瓜发酵多糖能够显著调节与肥胖相关的脂类代谢,比如游离脂肪酸(FFA)。结论:苦瓜发酵与未发酵多糖均能改善肥胖大鼠的高血脂症,而发酵多糖相比于未发酵多糖具有更显著抗高血脂作用的原因可能是由于其发酵作用。

【Abstract】 Objective:This research was aimed to investigate the effect of polysaccharides(FP) from fermented Moniordica charantia by Lactobacillus plantarum NCU116 on serum lipids and its mechanism in obese rats induced by high fat diet.Methods:Totally 50 male rats were randomly divided into 4 groups:(1) Normal,fed normal diet;(2) Model,fed high fat diet;(3) FP,fed high fat diet with FP.(4) NFP,fed high fat diet with NFP.Oral administrations were conducted with 10 mL/kg of b.w.over the 8-week experimental period in obese rats.Body weight and physiological condition was recorded daily.Levels of serum lipids including total cholesterol(TC),triacylglycerols(TG),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C) were determined;Serum insulin,leptin,and adiponectin were estimated using the assay kits;The pathological section of fat tissue was analyzed;Lipidomics was investigated by UPLC-Q-TOF-MS analysis.Results:FP could effectively lower the body weight gain and cell sizes of epididymal adipose tissues in obese rats comparing with polysaccharides from non-fermented Moniordica charantia(NFP);Furthermore,FP treatment on rats could significantly decrease serum TC,TG,and LDL-c,leptin,and increase serum HDL-c,adiponectin.Additionally,lipidomics results indicated that the lipids-associated with obesity(such as FFA) were significantly altered by FP treatments.Conclusion:both FP and NFP could improve hyperlipidemia in obese rats.FP exerted more effective impact on hyperlipidemia than NFP,which may attribute to fermentation.
