【摘要】 目的:对鳕鱼鱼鳔来源酶解肽的抗氧化活性与延缓H2O2诱导2BS细胞衰老作用的研究。方法:利用复合蛋白酶对鳕鱼鱼鳔进行酶解,将得到的酶解产物进行G-15葡聚糖凝胶柱层析。对分离纯化组分进行傅里叶红外光谱扫描、紫外光谱扫描、氨基酸组成分析与分子量测定。检测鱼鳔肽对DPPH·,HO·与O2·-自由基体外清除能力与Fe2+螯合能力。通过H2O2诱导2BS细胞建立早期细胞衰老模型,对鱼鳔肽对氧化诱导衰老细胞的细胞增殖率、ROS活性氧含量、β-半乳糖苷酶含量与细胞凋亡率进行检测。结果:通过分离纯化得到两个组分,分别命名为SWP-Ⅰ与SWP-Ⅱ。体外清除自由基能力实验结果显示两个组分对DPPH·,HO·与O2·-自由基具有一定的清除能力,且具有较好的Fe2+螯合能力。对H2O2诱导的早期衰老2BS细胞,鳕鱼鱼鳔肽能够有效提高细胞增殖率,降低ROS活性氧含量、β-半乳糖苷酶含量与细胞凋亡率。结论:通过酶解得到的鳕鱼鱼鳔肽具有较好的抗氧化能力并对H2O2诱导的早期衰老2BS细胞具有一定的保护作用,能够有效地延缓细胞衰老。

【Abstract】 Objective:Antioxidant activity of enzymatic peptides from Swimming bladder of codfish and its anti-aging effects on 2 BS induced by H2O2.Methods:Swimming bladder peptides were obtained by enzymatic hydrolysis with compound protease and then isolated by chromatographic method with G-15.The fractions were characterized by FT-IR,UV scanning spectrum amino acid composition analysis and molecular weight determination.The antioxidant such as scavenging activities on DPPH ·,HO · and O2 ·- and chelating activity of Fe2+ of peptides were evaluated in vitro.The cell viability,intracellular reactive oxygen species production,content of β-galactosidase and apoptosis rate in human fetal lung diploid fibroblast cells induced by H2O2 were detected.Results:Two bioactive peptides(SWP-Ⅰ and SWP-Ⅱ) were isolated by chromatographic method with G-15.The results of free radical scavenging showed that SWP-Ⅰ and SWP-Ⅱ possessed good scavenging activities on DPPH ·,HO · and O2 ·- and good Fe2+ chelating activity.Both of two peptides reduced intracellular reactive oxygen species production,content of β-galactosidase and apoptosis rate in human fetal lung diploid fibroblast cells induced by H2O2.Conclusion:The enzymatic peptides from swimming bladder of codfish possessed antioxidant activities and could protect the early aging cells induced by H2O2 effectively.
