植物乳杆菌素的基因组学及其结构研究

【摘要】 植物乳杆菌素因其无毒、高效,可作为食品生物防腐剂。荧光假单胞菌为食品中主要嗜冷污染菌之一,本课题组前期研究得到的植物乳杆菌细菌素对其有明显抑制作用。本研究从基因组学及结构两方面对植物乳杆菌素进行研究。对Lactobacillus plantarum Q7产生的细菌素基因组学进行研究,结果表明L.plantarum Q7含有1个完整的环化染色体和4个完整的环化质粒,GC含量为44.93%,染色体和质粒大小分别为2.95 Mb,12.46,2.96,2.41,11.89kb。通过蛋白质编码基因比对,显示在质粒pLP-Q704中存在细菌素ABC转运基因、细菌素转运辅助蛋白、细菌素免疫基因和细菌素编码基因,通过翻译细菌素编码基因,得到细菌素氨基酸序列;利用软件对细菌素的一、二级结构进行预测分析,其二级结构为α-螺旋(6.28%),β-折叠(20.45%)和无规则卷曲(72.73%)。本研究为植物乳杆菌素的合成调控与高效表达研究奠定了基础。

【Abstract】 Lactobacillus bactericin as a biological preservative can improve food quality and safety because of its non-toxic and high efficiency.Pseudomonas fluorescens is one of the main psychrophilic bacteria in food and the bactericin produced by Lactobacillus plantarum had a significant inhibitory effect on Pseudomonas fluorescens in previous study.This study was conducted from the whole genome sequence of producer strain and the structure of bactericin.According to the method of whole genome sequencing,the genome contained a circular chromosome(2 952 198 bp),four circular plasmids(12 458,2 968,2411 bp,and 11 899 bp,respectively),and GC content of 44.93%.According to Blast program,the gene cluster of bactericin was found in the pLP-Q704,including the bactericin ABC transport protein gene,the bactericin helper protein,the bactericin immunity protein and the bactericin encoding gene.The bactericin amino acid sequence was obtained by translating the bactericin-encoding gene.The primary and secondary structures of bactericin were analyzed by software,and the predicted secondary structure contained helix(6.28%),strand(20.45%) and loop(72.73%).This study laid the foundation for the synthesis regulation and high-efficiency expression of Lactobacillus plantarum Q7.
