基于共敏化结构的光电化学传感器用于凋亡细胞的灵敏检测

【摘要】 光电化学分析法具有灵敏度高、设备简单、成本低等的优点,吸引了科研工作者广泛的研究兴趣。作为一种检测生物标志物的分析技术,其在DNA分析、免疫分析、酶促传感以及细胞相关分析上具有良好的应用前景。本工作首先制备了还原氧化石墨烯(RGO)、ZnIn2S4纳米晶共敏化空心TiO2的光电复合电极,随后,以此复合电极作为基底构建了光电化学传感器用于检测凋亡细胞,并对白血病药物疗效进行评估。首先合成空心TiO2纳米球,在空心TiO2表面利用电沉积镀上电还原氧化石墨烯,提供电极的生物相容性和导电性,并利用连续离子层吸附与反应技术在其表面沉积ZnIn2S4纳米晶,形成光电化学基底电极,用于固定多肽以捕获凋亡细胞,通过光电流信号的变化间接测量细胞的浓度,并以此评估药物治疗细胞的疗效,结果表明药物能诱导细胞凋亡。此传感平台具有稳定性好、生物相容性好及可重复性好等特点,可拓展至其他抗癌药物的疗效评估。
【Abstract】 Photoelectrochemical bioanalysis has attracted a wide range of research interests because of its high sensitivity, simple equipment and low cost.Herein, photoelectrochemical cell sensor based on hollow TiO2/RGO/ZnIn2S4 cosensitized structure was developed for the detection of apoptotic cells and evaluation of drug effects.Hollow TiO2 nanospheres were non-toxic and had good light stability, which is suitable for cell sensing platform.The electrodeposition of graphene oxide on the surface of hollow-TiO2 photoelectrode provides biocompatibility and conductivity.ZnIn2S4 could be developed by the use of continuous ion-layer adsorption and reaction technology.The peptides could be immobilized to capture the apoptotic cells on the photoelectrochemical substrate electrode.The apoptotic cells could be detected indirectly by the change of the photocurrent signal.The platform has the characteristics of good biocompatibility and good reproducibility, which can also be extended to the evaluation of other anticancer drugs.
