新型免疫层析分析方法的研究

【Abstract】 The convention immunochromatographic assay(ICA) based on colloidal gold nanoparticles displays the properties of simple operation,short assay time and low assay cost.However,the normal ICA can only be used for qualitative or semi-quantitative analysis.In the last years,with the development of other nanoparticles such as quantum dots(QDs) and the noble metal nanoparticles with the high effect of surface-enhanced Raman spectroscopy(SERS),the research on sensitive and quantitative ICAs has been put forward greatly.Herein we report two sensitive and quantitative ICAs.(1) Fluorescent ICA(FICA).The oily-phase QDs with strong fluorescent emission and high quantum yield were firstly synthesized and then transferred to water-soluble QDs by enwrapping oily-phase QDswith polymer.The fluorescent immune-probe was formed by immobilizing the antibody against target analyte on the surface of the QDs microsphere.Using this probe,the FICA for the detection of brombuterol,a new emerged β-agonist,in food samples was developed.The sensitivity(IC50 value) of the FICA was 0.014 ng mL-1 and the limit of detection(LOD) was 0.53 pg mL-1.(2) SERS based ICA(SERS-ICA).Au particles were synthesized and then on the surface of the Au particles,Raman active species(4-mercaptobenzoic acid,MBA) were immobilized.Using MBA immobilized Au particles as a core,a silver shell with higher SERS effect was capped on the core to form AuMBA@Ag core/shell nanoparticles.The AuMBA@Ag nanoparticles were linked to antibody against target analyte to produce the SERS immune-probe AuMBA@Ag-Ab.Based on this probe,the ultrasensitive SERS-ICA for the detection of salbutamol in food sample was developed.The sensitivity for salbutamol was 0.027 ng mL-1 and the limit of detection(LOD) was 2.8 pg mL-1.Both ICAs exhibit the features of high sensitivity and specificity,simple operation,short assay time(<30 min.) and capable on site analysis.It is expected that using competitive immunoassay format,these ICAs can also be applied for other toxic substances or contaminants in food analysis.
