Norditerpenoids and sesquiterpenoids from the leaves of Crataegus pinnatifida(山楂)with antithrombotic evaluation

【摘要】 The leaves of Crataegus pinnatifida(Rosaceae),a well-known Traditional Chinese Medicine,have been used to cardiovascular for several decades[1,2].For a long time,its flavonoid was considered as the only effective component until small terpenoids with antithrombotic activity were obtained recently from hawthorn leaves[3-5].As a new effective ingredient,small terpenoids exhibit potential antithrombotic activity both in vitro and in vivo[6,7].Further work on characteristics of small terpenoids,two novel norditerpenoids with unprecedented carbon skeletons were isolated as the first diterpenoids from leaves of the title plant,together with two new sesquiterpenoids(3,4) and two known ones(5,6).The absolute configurations of 1-6 were determined by producing of Rh2(OCOCF3)4 complexes or Mosher ester,and by comparison of ECD,NMR,and optical rotation(OR) data to the calculated values.In this report,the isolation,structure elucidation,and in vitro and in vivo antithrombotic evaluation of all the isolates are described.This is the first time to obtain the norditerpenoids from leaves of the title plant.Once again,it enriched the kinds of natural products in hawthorn.Moreover,two norditerpenoids and four sesquiterpenoids were evaluated using multi-antithrombotic assays,showing significant activities both in vitro and in vivo.Meanwhile,the cytotoxicity studies indicated that all the isolates were nontoxic on three different cell lines.In aggregation,the extract of C.pinnatifida,as the resource of small terpenoids,were prepared according to the clinical medicine Yixintong.Although they may not exist abundantly as well as high flavonoid content,it still prompt that small terpenoids could play an important role solely or in good synergy with flavonoids.That need to be examined in future reseach studies.
