荧光双光子共聚焦与UPLC-HRMS法探究香草硫缩病醚在烟草植株中的内吸传导特性

【摘要】 [目的]探究香草硫缩病醚在烟草植株中的内吸传导特性,为其在田间科学合理用药方式提供参考。[方法]通过水培法和叶面喷雾法处理烟草植株,使用超高效液相色谱串联高分辨质谱(UPLC-HRMS)和荧光双光子共聚焦显微镜分别考察2种不同处理方式下香草硫缩病醚在烟草植株中的内吸传导性。[结果]采用水培法处理烟草植株根部时,香草硫缩病醚能够内吸向上传导至茎叶部位,12 h后茎叶样品中香草硫缩病醚含量达到最大值31.95 mg/kg;采用叶面喷雾法处理烟草植株叶面时,香草硫缩病醚能够内吸向下传导至根部位,72h后根部样品中香草硫缩病醚含量达到最大值0.215 mg/kg。此外,经过荧光双光子共聚焦成像,能够实时地观察到香草硫缩病醚在烟草的根、茎叶中内吸传导,与高分辨质谱定性定量的结果一致,进一步验证了香草硫缩病醚在烟草植株中具有内吸传导性。[结论]试验结果表明香草硫缩病醚在烟草植株中具有较好的向上和向下内吸传导性能,该项研究有助于指导香草硫缩病醚在田间科学合理的用药方式提供一定的参考。
【Abstract】 [Aims]The purposes of this study were to explore the systemic properties of XCLSBM in tobacco plant and to provide a reference for the rational use of XCLSBM in the field.[Methods]After the tobacco plants were treated by hydroponics and foliar spraying,The content of XCLSBM in root and stem leaf were detected by high performance liquid chromatography tandem high resolution mass spectrometry(UPLC-HRMS),the position of XCLSBM in root and stem leaf were real-time observed through fluorescence two-photon confocal microscope.[Results]XCLSBM was detected in the stem leaf were treated by hydroponics,with the extension of processing time,the content gradually increased.After 12 h,the content of XCLSBM is maximum,31.95 mg/kg.XCLSBM was detected in the root were treated by foliar spraying,with the extension of processing time,the content gradually increased.After 72 h,the content of XCLSBM is maximum,0.215 mg/kg.In addition,through the two-photon confocal fluorescence imaging,XCLSBM in real-time observed in the root and stem of tobacco leaf.This result was further shown the XCLSBM have systemic in tobacco plants.[Conclusions]The results showed XCLSBM has excellent upward and downward of systemic in tobacco plants,which is helpful to guide a reference for the rational use of XCLSBM in the field.
