桑树钾转运体MaHAK5基因的克隆与表达分析

【摘要】 钾(K~+)是植物生长发育所必需的大量元素之一,而且也是唯一一种植物所需的以较高浓度存在的阳离子。为了应对低钾胁迫,植物进化出一系列复杂的信号调控途径。HAK5是重要的高亲和钾转运体,它主要负责低钾条件下的钾吸收,低钾胁迫可以显著诱导HAK5的转录升高。本实验在桑品种育71-1的幼叶中克隆获得该基因的完整阅读框(ORF),命名为Ma HAK5。序列分析显示:Ma HAK5长2625 bp,ORF长2478 bp,编码825个氨基酸残基,预测编码的蛋白质分子质量为92.17k D,等电点为8.37。基因序列系统进化分析表明:育71-1与川桑、枣、森林草莓的亲缘关系较近。干旱、高盐和低温胁迫会影响桑树Ma HAK5基因的表达水平,低钾处理下Ma HAK5基因的表达水平升高并且影响生理生化指标。
【Abstract】 Potassium(K +) is one of the many elements necessary for plant growth and development,and is the only one that plants need to have a higher concentration of cation.In order to cope with the low potassium stress,the plant evolves a series of complex signal regulation pathways.HAK5 is an important high-affinity potassium transporter,which is mainly responsible for potassium uptake under low potassium conditions,and low potassium stress can significantly induce transcription of HAK5.In this experiment,the complete reading frame(ORF) of the gene was cloned from the young leaves of mulberry Yu 71-1,named MaHAK5.Sequence analysis showed that the MaHAK5 is 2625 bp long,It’s open reading frame(ORF) is of 2478 bp,and encoding 828 amino acids with a predicted molecular weight of 92.17 kD and an isoelectric point of 8.37.Phylogenetic analysis based on MaHAK5 with other species showed that mulberry had a closer relationship with Chuansang(Morus notabilis XP010112002.1),Jujube(Ziziphus jujuba XP015898221.1),Fragaria vesca(Fragaria vesca subsp.Vesca XP004299195.2).Drought,high salt and low temperature stress could affect the expression level of MaHAK5 gene in mulberry,and the expression level of MaHAK5 gene was increased and the physiological and biochemical indicators were affected by low potassium treatment.
