华根霉全细胞脂肪酶酯合成动力学拆分2-辛醇

【摘要】 <正>华根霉CCTCC M201021全细胞脂肪酶在非水相中具有较强的酯合成能力,是一种极具价值的生物催化剂。本研究进一步考察该全细胞脂肪酶的光学选择性应用,发现该脂肪酶在有机相中对酯合成动力学拆分外消旋2-辛醇具有较好的光学选择性(E>100),对其他手性砌块仲醇1-苯乙醇也具有较好的拆分能力。研究选择确定了合适的酰基供体和反应溶剂。进一步研究发现体系水活度的减少对反应的光学选择性没有明显影响,但对反应起始初速度有显著的提高作用。在相同转化率下,3A分子筛的添加使反应起始初速度提高了6.33倍。通过与其他4种商品化脂肪酶比较表明,在相同条件下华根霉全细胞脂肪酶对2-辛醇的拆分不但具有较好的光学选择性(E=102.58),而且也表现出较高的反应速率和转化率。综合研究结果充分显示了华根霉全细胞脂肪酶在非水相生物催化拆分应用中具有良好的应用前景。

【Abstract】 Whole-cell lipase from Rhizopus Chinensis CCTCC M201021 was proved to be a useful biocatalyst for esters production in organic solvent.Application of the whole-cell lipase on enantioselectivity had been studied.The result showed that there was highly enantzoselectivity(E>100) during the kinetic resolution of 2-octanol by esterification catalyzed by lipase in nonaqueous media.Moreover,the whole-cell lipase could also well enantioselective esterify other 2-alkanols like 1-phenylethanol which serve as chiral building blocks.In addition,appropriate acyl donors and solvents had been chosen.It was found that water activity did not obviously affect the enantioselectivity of lipase but could notable improve the rate of the reaction.At the same conversion,the initial rate of production of 2-octyl caprylate which added with 3 A molecular sieves was 7.33 times than without 3A molecular sieves.At last,compared with four commercial lipases at the same condition,the whole-cell lipase of Rhizopus chinensis CCTCC M201021 showed not only high enantioselectivity(E=102.58) but also more agreeable initial rate and conversion.The results obtained in this study demonstrated the applicability of the whole cell biocatalyst to enantioselective biocatalysis in nonaqueous media.
