钙添加对镉污染水体草鱼的保护及其机制
Protective effect and mechanism of calcium addition on grass carp in cadmium-polluted water

【摘要】 本研究旨在分析低浓度镉(0.01mg/L)污染水体中添加钙对草鱼的保护作用及其机制。实验设置对照(0 mg/L)、低钙(30 mg/L)、中钙(60 mg/L)和高钙(90 mg/L)四个组,历时30天。结果显示:添加Ca2+对草鱼生长并无显著影响(p>0.05)。所有实验组草鱼组织Cd富集量顺序均为肾>肠>鳃>肝>肌肉≈血液,且肾脏和鳃Cd富集量随Ca2+浓度的增加呈显著下降趋势(p<0.05)。Ca2+显著降低了草鱼血浆Na+和Cl-浓度。中钙组草鱼肝脏POD、CAT和T-SOD活性均显著低于其他实验组(p<0.05),高钙组草鱼肝脏MDA含量显著高于其他组(p<0.05);中钙组和高钙组草鱼肌肉中POD、CAT、T-SOD活性均显著低于其他实验组(p<0.05),MDA含量无显著差异(p>0.05)。鳃Na+/K+-ATPase、Ca2+-ATPase活性变化在各组间的差异变化显著(p<0.05)。高钙组草鱼鳃Na+/K+-ATPase和Ca2+-ATPase基因表达量显著高于其他实验组(p<0.05)。研究结果表明,外源钙添加对镉污染水体草鱼具有一定的保护作用。
【Abstract】 In order to analyze the protective effects and mechanism of Ca2+ on Cd toxicology in grass carp(Ctenopharyngodon idella) in water polluted with low Cd(0.01 mg/L). The experiment was conducted in four groups with control(0 mg/L), low(30 mg/L), medium(60 mg/L), and high Ca2+(90 mg/L) for 30 days. The results showed that different concentrations of Ca2+ had no significant effect on grass carp growth(p > 0.05). The Cd bioaccumulation level in all examined tissues were kidney>intestinal>gill>liver>muscle≈blood. The Cd concentration in kidney and gill decreased significantly with the increase of Ca2+ concentration(p<0.05), and Ca2+ significantly reduced the plasma Na+ and Cl-concentrations(p<0.05). The activities of POD, CAT and T-SOD in the liver of the medium Ca group were significantly lower than those in other groups(p<0.05), and the MDA content in the liver of the high Ca2+ group was significantly higher than that in other groups(p<0.05). The activities of POD, CAT and T-SOD in the muscles of the middle Ca2+ group and the MDA contents in muscle of high Ca2+ group were significantly higher than other groups(p<0.05). The activities of Na+/K+-ATPase and Ca2+-ATPase in the gill of grass carp was significantly different among treatments(p<0.05). The m RNA expression levels of gill Na+/K+-ATPase and Ca2+-ATPase genes were the highest in high Ca2+ group. The results showed that the addition of exogenous Ca had a certain protective effects on the toxicity of Cd to grass carp.
