使用转录组学和蛋白质组学方法联合分析大黄素对嗜水气单胞菌的抑制作用

【摘要】 为探讨大黄素对嗜水气单胞菌抑菌作用机制,使用RNA-Seq和i TRAQ技术联合检测并比较分析了正常组和200μg/ml大黄素组对嗜水气单胞菌的差异基因及蛋白质表达谱变化。结果显示:获得158个基因(88个上调,70个下调)和237个蛋白(52个上调,185个下调)具有差异表达,关联分析表明81个基因和蛋白表达一致(64个上调,17个下调)。GO功能注释表明,显著差异基因主要参与翻译、ATP合成耦合质子输运、硫化氢生物合成过程、甘油分解等过程。KEGG富集分析表明,显著差异基因主要参与核糖体和氧化磷酸化等通路。其中有差异的前10个基因或蛋白分别是:AHA2481、hgpB、AHA0893、hscB、AHA1848、yfiA-2、napA、AHA1844、AHA2996和AHA1359等蛋白在转录和蛋白水平共表达,主要参与了毒力、细胞膜、细胞质合成、能量代谢等过程。因此,大黄素可能通过改变嗜水气单胞菌相关的毒力基因、细胞膜调控蛋白和能量代谢蛋白等抑制嗜水气单胞菌的生长。
【Abstract】 In order to investigate the antibacterial mechanism of emodin on Aeromonas hydrophila, RNA-Seq and i TRAQ techniques were used to detect and compare the differential genes and proteins of A. hydrophila in normal group and 200μg/ml emodin group. Expression changes. The results showed that 81 genes and protein expression were consistent(64 up-regulated, 17 Downgrade). GO function annotation indicates that significant differential genes are mainly involved in translation and glycerol decomposition etc. KEGG enrichment analysis showed that significantly different genes are mainly involved in pathways such as ribosomes. The top 10 genes or proteins are: AHA2481、hgpB、AHA0893、hscB、AHA1848、yfiA-2、napA、AHA1844、AHA2996 and AHA1359 are co-expressed at the transcriptional and protein levels, mainly involved in virulence, Processes such as cell membrane, cytoplasmic synthesis, and energy metabolism. Therefore, emodin may inhibit the growth of A. hydrophila by altering the virulence genes and cell membrane regulatory proteins.
