饲料组氨酸水平对团头鲂幼鱼的肠道抗氧化酶活和基因表达的影响

【摘要】 制作六组不同梯度组氨酸水平的半纯化饲料(从4.1到14.2 g kg-1)喂鱼8周。结果表明,与对照组相比,肠道丙二醛(MDA)、总超氧化物歧化酶(T-SOD)和谷胱甘肽过氧化物酶(GPx)的活性在组氨酸水平8.0 g kg-1明显较低。铜锌超氧化物歧化酶(Cu/Zn-SOD)和锰超氧化物歧化酶(Mn-SOD)的活性在组氨酸水平9.9 g kg-1最低。总抗氧化能力(T-AOC)和过氧化氢酶(CAT)的活性在组氨酸水平14.2 g kg-1明显高于8.0和9.9 g kg-1水平。饲料组氨酸水平对血液生化指标没有显著影响。Kelch样环氧氯丙烷相关蛋白-1(Keap1)mRNA水平在组氨酸水平8.0 g kg-1提高,而抑制了核因子E2相关因子2(Nrf2)mRNA水平,随后减少了CAT、GPx1、Cu/Zn-SOD和Mn-SOD相对表达水平。这些结果表明,饲料组氨酸在维持团头鲂幼鱼的肠道健康方面起着重要作用。

【Abstract】 Six semi-purified diets of graded histidine levels(from 4.1 to 14.2 g kg-1) were fed to fish for 8 weeks. The results showed that the intestinal content of malondialdehyde(MDA), total superoxide dismutase(T-SOD) and glutathione peroxidase(GPx) were noticeably lower in 8.0 g kg-1 diet compared to control group. While the lowest activities of Copper zinc superoxide dismutase(Cu/Zn-SOD) and manganese superoxide dismutase(Mn-SOD) were observed in 9.9 g kg-1 diet. The intestinal activities of total antioxidant capacity(T-AOC) and catalase(CAT) in 14.2 g kg-1 diet were noticeably higher than those in 8.0 and 9.9 g kg-1 diets. Plasma biochemical indexes were not significantly affected by dietary histidine levels. The Kelch-like-ECH-associated protein 1(Keap1) mRNA levels were increased in 8.0 g kg-1 diet, which suppressed the nuclear translocation of nuclear factor E2-related factor 2(Nrf2), and subsequently decreased CAT, GPx1, Cu/Zn-SOD and Mn-SOD expression levels In the intestine. These results indicated that dietary histidine plays a major role in maintaining intestinal health in juvenile blunt snout bream.
