斑节对虾耐低盐相关基因的克隆、表达分析及关联SNP的开发

【摘要】 为获得可靠的斑节对虾耐低盐性状分子标记,应用于斑节对虾分子标记辅助育种,本研究通过末端快速扩增(RACE)方法,从斑节对虾中获得了葡萄糖转运蛋白基因1的全长。利用直接测序法,共筛得28个SNP候选位点。斑节对虾葡萄糖转运蛋白1氨基酸序列与其他无脊椎动物的相似性为72%-99%。RT-PCR结果显示该基因在各组织中广泛表达,在鳃和肝胰腺中表达量最高,中等程度表达于心脏、肠、胃、眼柄神经和淋巴,在肌肉、精巢和卵巢中表达量较低。在盐度3ppt胁迫下,在鳃、肝胰腺中葡萄糖转运蛋白1的m RNA水平均出现了不同程度的变化。

【Abstract】 Reliable molecular markers of low salinity traits was obtained to applying to molecular marker-assisted breeding of Penaeus monodon. In this study, the full length of glucose transporter 1 was cloned by rapid amplification of cDNA ends( RACE). A total of 28 SNPs candidate sites were screened by direct sequencing. Similarity analysis showed that the amino acid sequence of the glucose transporter 1 of P. monodon shared high identity with other invertebrates by 72%-99%. Real-time PCR results showed the expression of this gene in all tested tissues. The results indicated that the highest expression levels were detected in gill and hepatopancreas, moderate expression in heart, intestine, stomach, eyestalk nerve and lymph, whereas the lowest expression levels were detected in muscle, testicle and ootheca. The mRNA level of glucose transporter 1 in gill and hepatopancreas changed significantly, after 3 ppt salinity stress.
