大黄素介导ABC转运蛋白暴露二嗪农下在草鱼组织研究

【摘要】 为研究ABC转运蛋白受中药提取物大黄素介导,同时暴露于二嗪农,ABC家族基因在草鱼不同组织表达差异。通过检测ABC亚型ABCB1,ABCB11,ABCC1,ABCC2和ABCG2基因在肝脏,肠道,肌肉,肾脏,脾脏,心脏,脑,鳍条,皮肤,鳃组织表达丰度。结果显示ABC家族基因主要表达于肝脏,肠道,鳃和皮肤。同时检测了在大黄素介导暴露二嗪农,检测肝脏和肠0.5 h,1 h,3 h,6 h,12 h,24 h,2 d,3 d,5 d和7 d药物处理后不同时间点ABC家族基因表达特征,显示肝脏中ABCB1,ABCB11,ABCC1,ABCC2和ABCG2最高表达丰度在暴露后24h;而肠道中ABCB1和ABCB11最高表达与暴露后6h,ABCC1和ABCC2丰度最高于暴露后5 d,ABCG2丰度最高于暴露后3 d。结果显示出大黄素介导二嗪农暴露下,不同ABC基因亚型之间,其表达时间和表达丰度具有不一致性,且ABCG2表达丰度高于其他亚型,由此判断大黄素介导二嗪农的过程主要经由ABCG2参与转运。同时通过LC-MS-MS检测肝脏和肠道组织大黄素和二嗪农蓄积浓度,显示大黄素在肝脏中最高浓度(50.18μg·ml-1)和肠道(117.24μg·ml-1)分别在给药后48 h和1 h;二嗪农浓度在肝脏(1.42 ng·ml-1)和肠道(0.2 ng·ml-1)分别在药物处理后3 h和6 h。显示大黄素和二嗪农蓄积时间点和浓度在肝脏和肠道中差异显著。

【Abstract】 The diazinon(DZN) as an organophosphorus pesticides was polluted the aquaculture. In order to investigate the ABC regulated by emodin then transport to DZN in grass carp. The ABC transporter genes were measured in tissues and different time points in grass carp treated with emodin and exposed to DZN. The ABCB1, ABCB11, ABCC1, ABCC2 and ABCG2 genes were highly expressed at 24 h in liver in the experimental group. However, in intestinal tissue, the ABCB1 and ABCB11 genes exhibited peak expression at 6 h, the ABCC1 and ABCC2 genes exhibited peak expression at 5 d, and the ABCG2 gene exhibited peak expression at 3 d in the experimental group. In this study, the expression of ABCG2 was higher than that of the other ABC transporter genes, which showed that ABCG2 may play an important role in transport processes in grass carp treated with emodin and exposed to DZN. Furthermore, the concentrations of emodin and DZN were measured. In the different time points, the emodin concentrations peaked in liver(50.18 μg·ml-1) 48 h and intestine(117.24 μg·ml-1) 1 h respectively. The DZN concentrations peaked in liver(1.42 ng·ml-1) and intestine(0.2 ng·ml-1) after 3 h and 6 h of DZN exposure, respectively.
