宽礁膜游孢子萌发及个体发生过程的初步观察

【摘要】 宽礁膜(Monostroma Latissimum)生活史分为叶状体和囊状孢子体两个阶段,孢子体成熟后放散游孢子萌发形成叶状体。本实验基于宽礁膜异形世代交替的生活史,研究了游孢子放散固着后初期个体发生的过程,包括细胞分裂、假根形成、叶面积比增长率等。结果表明:1)游孢子完成固着后,细胞极性特征明显,即一端完成胞内物质的积累与分裂,另一端延伸出无色假根;2)其萌发过程通过纵向分裂和横向分裂交替完成;3)游孢子个体发生的三种细胞形态,假根细胞、基部细胞、营养细胞;4)假根形成与附着基质有关,同期固着在筛娟上的游孢子假根与玻璃盖玻片上差异显著(P<0.05);5)单个游孢子可萌发成单株苗或多株苗。6)随着细胞分裂加快,叶面积比增长率提高,并呈指数增长。本实验对游孢子萌发及个体发生的基础性研究为宽礁膜产业化栽培提供数据支撑。

【Abstract】 The life history of Monostroma Latissimum is devided into two stages: thallus and cysts. Zoospores are released after cysts’ s maturation, then form thallus after attachment. Based on the life history of alternation of heteromorphic generations, the process of ontogeny was studied, include cell division, rhizoid formation and specific growth rate of thallus area et.al. The results show that: 1) The characteristic of polarized cells is obvious, one side completed the accumulation of Material and cell division,and another side extended rhizoid. 2) The germination and ontogeny finished by vertical-division and transverse-division. 3) There are three types of cell morphology, including rhizoid cells, basal cells and nutrient cells. 4) Rhizoid formation was related to the attachment substrate, it is significant between screen and glass cover(P<0.05). 5) Single zoospore can germinate into single or multiple seedlings. 6) With the acceleration of cells division, the thallus area increased exponentially. This study for basic research on zoospores’ s germination and ontogeny provides data support for the industrialization cultivation of Monostroma Latissimum.
