江苏省河蟹肝孢虫流行病学调查及RPA检测方法建立

【摘要】 微孢子虫是一类广泛分布于自然界专营细胞内寄生的真核原生动物,是许多具有经济价值的昆虫、甲壳类、鱼类、啮齿类等动物的病原体。在2007年发现一种只寄生于中华绒螯蟹肝胰腺上的微孢子虫命名为河蟹肝孢虫(Hepatospora eriocheir)。为了解江苏省部分养殖区河蟹肝孢虫的感染情况,本文首先采用PCR技术对采集的河蟹肝胰腺进行检测。同时,建立了河蟹肝孢虫的重组酶聚合酶等温扩增(RPA)检测方法。结果显示:2017年在调查的江苏省5个地区中,只有3个地区检测出河蟹肝孢虫,其中常州前黄的感染率最高,其次为南通和兴化安丰;2018年在调查的江苏省4个地区中,有2个地区检测出河蟹肝孢虫,其中扬州高邮的感染率最高,其次是无锡宜兴;此外,建立RPA检测方法能够检测到376 bp的特异性条带,仅需在37℃下恒温反应30min,该检测方法扩增时间短、效率高、对设备的要求低,适合于基层简易实验的快速检测。

【Abstract】 Microsporidia is a kind of eukaryotic protozoa widely distributed in nature. It is the pathogen of many economic insects, crustaceans, fish, rodents and other animals. A microsporidia isolated from the hepatopancreas of Eriocheir sinensis was found in 2007 and named hepatospora eriocheir. In order to learn the infection status of Hepatospora eriocheir in Jiangsu Province, firstly, used PCR technology to detect the hepatopancreas. At the same time, the Recombinase Polymerase amplification(RPA) detection method of Hepatospora eriocheir was established. The results showed that only three of the five areas in Jiangsu Province were detected in 2017. The infection rate of Qianhuang in Changzhou was the highest, followed by Nantong and Xinghua Anfeng. In 2018, Hepatospora eriocheir was detected in two of the four regions in Jiangsu Province, of which Gaoyou in Yangzhou was the highest infection rate, followed by Yixing in Wuxi. In addition, a specific band of about 380 bp can be detected by RPA detection method, which only needs 30 minutes of isothermal reaction at 37℃. This method has the advantages of short amplification time, high efficiency and low equipment requirements, which is suitable for rapid detection of simple grass-roots experiments.
