斑节对虾C型凝集素抗菌作用及其在氨氮和盐度胁迫下表达模式的相关研究

【摘要】 C型凝集素作为模式识别受体(PRRs)超家族的一员,其通过可以特异性识别并结合病原相关分子模式(PAMPs)而在无脊椎动物先天免疫中发挥重要作用。斑节对虾(Penaeus monodon)C型凝集素含有一个CRD结构域。PmCLB在肝胰腺,心脏和淋巴特异性分布,卵巢中几乎不表达。当斑节对虾受到哈维氏弧菌(Vibrio harveyi)和鳗弧菌(Vibrio anguillaru)刺激时,PmCLB的m RNA水平在肝胰腺和鳃中的显著上升。PmCLB的重组蛋白(r PmCLB)能在体外结合脂多糖(Lipopolysaccharide,LPS),肽聚糖(Peptidoglycan,PGN)和甘露糖(mannose)等多种碳水化合物。r Pm CLB在钙离子的参与下能结合金黄葡萄球菌(Staphylococcus aureus),哈维氏弧菌(Vibrio harveyi)和鳗弧菌(Vibrio anguillarum)等多种细菌。r PmCLB能显著提高斑节血淋巴细胞对虾对哈维氏弧菌的清除效率。另外,当斑节对虾处于氨氮和低盐胁迫时,肝胰腺和肠中的PmCLB表达量有显著变化,以应对不利的环境。
【Abstract】 C-type lectins(CTLs),as a number pattern recognition receptors(PRRs), play important roles in invertebrate innate immunity by recognizing and eliminating pathogens. In the present study, C-type lectin-like domain(CTLD) containing CTL was characterize from the tiger shrimp Penaeus monodon, named as PmCLB. The full-length cDNA of PmCLB was composed of 861 bp, with a 495 bp ORF encoding a 165 aa protein. The mRNA transcripts of PmCLB was expressed the highest in hepatopancreas and hardly detected in ovary. After the shrimps were stimulated by Vibrio harveyi, Vibrio anguillarum, the expression level of PmCLB in hepatopancreas and gill were all significantly up-regulated. Carbohydrate binding assay revealed that the PmCLB had specificity to pathogen-associated molecular patterns(PAMPs),including PGN and LPS, and saccharides, such as mannose, D-glucose, galactosamine, lactose and treholose. The recombinant PmCLB agglutinates Gram-positive(Staphylococcus aureus) and Gramnegative bacterias(V.harveyi, V.anguillarum, Vibrio alginolyticus, Vibrio parahemolyticus, Vibrio vulnificus, and Aerononas hydrophila,) in the presence of calcium ion and also could enhance the efficiency of clearing invaded bacterias so that functioned as a PRR involved in the innate immunity of P. monodon.In addition, PmCLB could also response to adverse environment including ammonia nitrogen and salinity stress.
