母源免疫处理草鱼5种免疫因子含量变化规律初探

【摘要】 为探究不同剂量GCRV疫苗母源免疫草鱼血液中不同免疫因子的变化规律,本研究以一针免疫高剂量组、普通剂量组、低剂量组、二针免疫组和对照组的草鱼母本作为研究对象,采集草鱼母本产卵后的血液,运用ELISA技术测定5种免疫因子(Ig M、C3、LZM、MBL和Bf)的蛋白活性水平。结果显示经过GCRV疫苗处理的草鱼母本5种免疫因子的蛋白活性水平均高于对照组,其中一针免疫高剂量组5种免疫因子的蛋白活性水平为对照组的1.34-1.74倍,一针免疫低剂量组为对照组的1.04-1.26倍,一针普通剂量组为对照组的1.10-1.35倍,二针免疫组为对照组的1.00-1.16倍。本实验初步证明了通过GCRV疫苗刺激可以增强五种免疫因子的富集表达,不同剂量疫苗刺激能引起五种免疫因子产生不同的表达。该研究为阐明母源免疫处理草鱼免疫因子含量变化规律,深入了解草鱼母源免疫因子的抗病调控机理奠定了基础。
【Abstract】 To investigate the fluctuation of different immune factors in blood of Ctenopharyngodon idella immunized by different doses of GCRV vaccine, this study used a normal dose, a high dose, a low dose and two doses of immunization and the control group as the research objects. The protein activity levels of five immune factors(IgM, C3, LZM, MBL and Bf) were determined by ELISA. The results showed that the protein activity levels of the five immune factors of grass carp were significantly higher than those of the control group. The protein activity level of the five immune factors in the high-dose group was 1.34-1.74 times that of the control group, the low-dose group was 1.04-1.26 times that of the control group, the normal dose group was 1.10-1.35 times that of the control group, and the two-doses immunization group was 1.00-1.16 times of the control group. This experiment preliminarily proved that the stimulation of GCRV vaccine can enhance the enrichment expression of five immune factors, and different doses of vaccine stimulation can cause different expression of five immune factors. This study laid the foundation for elucidating the fluctuation of five immune factors in grass carp after maternal immune and understanding the mechanism of disease resistance regulation of grass carp maternal immune factors.
