载BMP-2多巴胺-肝素-透明质酸促进牙本质形成的研究

【摘要】 目的:探讨载BMP-2多巴胺-肝素-透明质酸支架材料对牙髓干细胞粘附牙本质、增殖、分化和形成牙本质的影响。方法:采用化学方法构建多巴胺-肝素-透明质酸,通过傅里叶变换红外光谱分析及核磁共振氢谱进行表征。应用扫描电镜观察其微观结构特点,测定其含水率和孔隙率。将骨形态发生蛋白-2(bone morphogenetic protein-2,BMP-2)载入多巴胺-肝素-透明质酸,测定BMP-2的释放情况和生物学活性。制备根管研究模型,以载有BMP-2的多巴胺-肝素-透明质酸构建牙本质表面复合支架,与牙髓干细胞共培养4,7和14天,扫描电镜观察细胞粘附、生长和增殖。结果:成功制备载BMP-2的多巴胺-肝素-透明质酸材料,其孔隙范围为130-180μm,可缓释BMP-2达3周,且能保持其生物学活性。结论:牙本质表面的载BMP-2多巴胺-肝素-透明质酸支架能够促进牙髓干细胞粘附牙本质、增殖、分化和形成牙本质。

【Abstract】 Objectives:To investigate the effect of dopamine-heparin-hyaluronic acid loaded with BMP-2 on the attachment,proliferation and differentiation of dental pulp stem cells(DPSCs) and dentin formation.Methods:The dopamine-heparin-hyaluronic acid was synthesized by chemical methods and characterized by FT-IR and ~1 HNMR.The water content and porosity were measured,and the microcosmic morphology was investigated by scanning electron microscope(SEM).Bone morphogenetic protein-2(BMP-2) was loaded into dopamine-heparin-hyaluronic acid,and then the release and biological activity of BMP-2 were measured.The root canal model were prepared and then the composite scaffold was fabricated on the dentin surface with BMP-2-loaded dopamine-heparinhyaluronic acid.DPSCs and dentin model coated with the composite scaffold were co-cultured for 4,7 and 14 days,and the adhesion,growth and proliferation of DPSCs were observed by SEM.Results:The dopamine-heparin-hyaluronic acid loaded with BMP-2 was successfully prepared,with the pore size of 130-180μm,which could sustained release BMP-2 with biological activity for 3 weeks.Conclusions:The dopamine-heparin-hyaluronic acid loaded with BMP-2 coating on the dentin surface promotes the attachment,proliferation and differentiation of DPSCs and dentin formation.
