连接桥粒斑株蛋白对口腔鳞癌HSC3细胞株增殖、迁移和侵袭影响的研究

【摘要】 目的本课题拟研究连接桥粒斑株蛋白(junction plakoglobin,JUP)对口腔鳞癌HSC3细胞株增殖、凋亡、迁移及侵袭的影响以及对OSCC患者预后生存质量的影响。方法 (1)用IHC检测274例OSCC组织中JUP表达水平,用卡方检验和Fisher确切概率法比较JUP表达高、低与患者临床病理特征的关系。采用Kaplan Meier和log-rank评价JUP表达与患者生存时间的关系,Cox回归模型对潜在影响预后的因素进行评估。(2)用WB及RT-PCR检测了JUP在正常口腔黏膜上皮细胞株NOK及人口腔鳞癌细胞株Ca127、HSC3、HSC4、HSC6、SCC25中的表达水平。(3)分别采用CCK8法、流式细胞术、Transwell小室检测过表达JUP对HSC3细胞增殖活性、凋亡、迁移和侵袭的影响。结果 (1)高表达JUP患者5年生存率均较低表达组低(GZ:p=0.006,<0.05;CD:p=0.016,<0.05),Cox回归模型显示高表达JUP是口腔鳞癌独立风险因素之一(GZ和CD危险比为1.97,2.27;95%置信区间分别为:p=0.012,p=0.003均小于0.05)。(2)WB、RT-PCR均显示Ca127、HSC3、HSC4、HSC6、SCC25口腔鳞癌细胞株较NOK组表达升高(P<0.05)。(3)过表达JUP促进HSC3细胞株的增殖、迁移与侵袭,而抑制了其凋亡。结论本研究初步证明JUP的高表达与OSCC患者的预后相关,且可作为一项独立危险评价因素。

【Abstract】 Objectives: The purpose of this study was to investigate the effects of junction plakoglobin(JUP) on proliferation, apoptosis, migration and invasion of human oral squamous cell carcinoma cell line HSC3 and its influence on the prognosis of patients with OSCC. Methods:(1)We detected the expression level of JUP in 274 OSCC specimens through immunohistochemistry, and the relationship of expression level of JUP in 274 clinical specimens with patients clinicopathologic characteristics was analysed by chi-square test and Fisher exact probability method. Kaplan-meier and log-rank were used to evaluate the relationship between JUP expression and patients survival time, and Cox regression model was used to assess the potential prognostic factors.(2) The expression levels of JUP in normal oral mucosal epithelial cell lines NOK and human oral squamous cell lines Ca127, HSC3, HSC4, HSC6, SCC25 were detected by WB and RT-PCR.(3) The effects of overexpression of JUP on the proliferation, apoptosis, migration and invasion of HSC3 cells were respectivelydetected by CCK8, Transwell and flow cytometry. Results(1) The five-year survival rate of patients with high expression of JUP was lower than the low-expression group(GZ: p=0.006, <0.05; CD: p=0.016, <0.05). Cox regression model showed that high expression of JUP was one of the independent risk factors for OSCC(GZ: Hazard ratio: 1.97, 95% confidence interval, p=0.012; CD: Hazard ratio: 2.27, 95% confidence interval, p=0.003).(2) WB and RT-PCR showed that the expression of cell lines Ca127, HSC3, HSC4, HSC6, SCC25 was higher than NOK group(P<0.05).(3) Overexpression of JUP promoted the proliferation, migration and invasion of HSC3 cell lines and inhibited their apoptosis. Conclusion: This study initial confirm that high expression of JUP was related to the prognosis of OSCC patients and can be an independent risk assessment factor.
