雌二醇酶联免疫吸附分析方法的建立

【摘要】 针对食品中雌二醇残留污染问题,本研究以雌二醇肟化后制备新型半抗原及系列人工抗原免疫动物后所取得的可特异性识别雌二醇的多克隆抗体为基础,建立了检测雌二醇的酶联免疫分析方法(ic-ELISA)。本方法对于雌二醇的半数抑制浓度(IC50)为5.55 ng/mL,线性检测范围为0.36~84.96 ng/mL,检出限为0.07 ng/mL;与激素类药物及结构类似物均无交叉反应,具有较好的特异性和灵敏度。
【Abstract】 To solve the problem of estradiol residual in foods, we deuterated the estradiol as a hapten and prepare artificial antigens to obtain polyclonal antibodies by immunization of animals. Based on the polyclonal antibodies, an enzyme-linked immunoassay(icELISA) for estradiol residues detection was established. For this method, the IC50 was 5.55 ng/mL and the linear detection range was 0.36-84.96 ng/mL with a detection limit of 0.07 ng/mL; the cross-reactivity with other hormone drugs and structural analogues was < 0.1%, showing the method had good specificity and sensitivity.
