切花菊‘粉贵人’高效再生体系的建立

【摘要】 ’粉贵人’是本课题组通过人工有性杂交获得的切花菊新品种,本研究以’粉贵人’叶片和茎间薄层为外植体,以MS为基本培养基,设立了不同浓度的6-BA和NAA组合,筛选出了适于两类外植体愈伤组织诱导和分化的最适培养基,其中叶片最适分化培养基为:MS+1mg/L 6-BA+1mg/L NAA,在此培养基上叶盘培养22d左右即可分化出不定芽芽点,40d时分化芽点既能生长成1cm左右小苗,60d时分化苗生根率既能达到100%,移栽后成活率达到100%;茎间薄层最适分化培养基为:MS+2mg/L 6-BA+1mg/L NAA,茎间薄层在28d左右分化出芽点,43d左右成苗;分化苗最佳生根培养基为1/2 MS+0.2mg/L NAA。为了针对’粉贵人’进行进一步的转基因研究,对两类外植体对卡那霉素的耐受性进行了分析,结果发现,叶片临界抗性选择压为5mg/L,茎间薄层临界抗性选择压为2mg/L,分化苗生根选择压为8mg/L。通过与其他报道对比发现,’粉贵人’是一个能够快速出愈、分化、生根的品种,且增殖系数高,再生过程中无需频繁更换培养基,是一个在生产和基因功能研究上均非常重要的切花菊新品种。
【Abstract】 ’Fenguiren’ is a new cut chrysanthemum species obtained through artificial hybridization.With leaves and tTCLs as explants,MS was used as the basic medium and it was supplemented with 6-BA and NAA at different concn for designing different media in order to study the optimum differentiation conditions of leaves and tTCLs.The optimum medium for leaves inducing adventitious bud differentiation were MS +2 mg/L 6-BA + 1 mg/L NAA,on which the explants were cultured to differential buds for about 22 days,the adventitious buds could grow to about 1 cm or so on the 40 th days,and the rooting rate of the differentiated shoots reached 100%at 60 days.While MS +2 mg/L 6-BA + lmg/L NAA was the optimum medium to differentiation for tTCLs,on which the tTCL-callus differentiated into buds at about 28 days,and adventitious buds grew to 1 cm about43 days.The best rooting medium for differentiation seedlings is 1/2 MS + 0.2 mg/L NAA.For further transgenic studies,the tolerance of kanamycin to the two types of explants was analyzed.The results showed that the critical resistance of the leaves was Smg/L,the tTCLs was 2 mg/L,and differentiation seedling rooting pressure is 8 mg/L Through comparison with the literature,it is found that the’ Fenguiren’ is a new cut chrysanthemum species that can quickly induce callus,differentiate,and take root,has a high proliferation coefficient,and does not need frequent replacement of the medium during regeneration,and is very important in both production and gene function studies.
