应用快速切片法观察芍药不同花型品种花芽分化进程
Observation of the Flower Bud Differentiation Process of Paeonia lactiflora by Using Rapid Section Method

【摘要】 为探索进行芍药花芽分化观察的高效方法,快速准确地确定芍药花芽分化进程,以芍药’粉玉奴’(单瓣型)、’朱砂判’(皇冠型)和’种生粉’(台阁型)地下芽为试验材料,对比分析石蜡切片的方法特点,采用快速切片法对其花芽分化过程进行了观察。结果表明:与石蜡切片法相比,快速切片法简易、省时、高效、环保,所获图谱质量可靠,适合作为芍药花芽分化过程形态观察研究。’粉玉奴’和’朱砂判’的花芽分化过程类似,均包括:苞片原基、萼片原基、花瓣原基、雄蕊原基、雌蕊原基等5个分化阶段,但’朱砂判’雄蕊原基分化后期出现离心瓣化。而’种生粉’花芽依次分化形成下位花、上位花(1)和上位花(2),下位花花芽分化过程包括:苞片原基、萼片原基、花瓣原基、雄蕊原基、雌蕊原基等5个分化阶段,上位花(1)花芽依次分化花瓣原基、雄蕊原基和雌蕊原基,而上位花(2)花芽仅观察到分化一轮花瓣原基。
【Abstract】 The morphological characteristics of flower buds during the differentiation process of flower buds were studied using the underground buds of ’Fenyunu’(single type),’Zhushapan’(crown type) and ’Zhongshengfen’(double-flowered type)by using rapid section method,after analysed the characteristics of the method of paraffin section method.Compared with the paraffin section method,the rapid section method is simple,time-saving,efficient,and environmentally-friendly.The quality of the obtained picture is reliable,and it is suitable as a morphological observation study of the flower bud differentiation process of the peony.The results showed that the flower bud differentiation processes of ’ Fengyunu’ and ’ Zhushapan’ were similar,and they all included five differentiation stages:sepal primordium differentiation stage,sepal primordium differentiation stage,petal primordium differentiation stage,stamen primordium differentiation stage,and pistil primordium differentiation stage.But the stamen petalody at the later stage of the development of ’ Zhushapan’.The flower buds of ’ Zhongshengfen’ differentiated in turn to form lower flower,upper flower(1) and upper flower(2).The differentiation process of the lower flower includes five parts:sepal primordium,sepal primordium,petal primordium,stamen primordia,and pistil primordia.And the upper(1) flower differentiated the petal primordium,the stamen primordium and the pistil primordium in turn,while the upper flower(2) only observed the differentiation of the simple layer of petal.The results provide important references for the research of flower bud differentiation of other plants with the similar morphological character of bud.
