【摘要】 目的:中医药具有整体观和辨证论治的特点,中药及其复方在防病治病过程中发挥较好的疗效。然而,其具有多成分、多靶点的特点,使中医药的现代化发展遇到困阻。因此,从中药复方中筛选有效的活性单体尤为重要。方法:近年来随着系统生物学的兴起,基于生物数据库的活性成分的筛选在国内外越来越受到关注。在这一背景下,本文在经典中药复方治疗疾病的启发下,提出一种寻找"活性单体"的新方法,即基于系统生物学的"成分-靶点-疾病"挖掘,采用多种数据库的全面的信息来寻找中药复方中针对疾病的关键靶点起治疗作用活性成分,实现活性成分与疾病靶点的对接,进而对单一有效成分进行实验验证。结果:通过该方法能够实现中医方剂筛选"活性单体",且通过与国外研究者从数据库获取的庞大数量的备筛选成分相比,从临床疗效明确的中药复方中寻找"活性单体",可大大缩小备筛选化合物的范围,而更具有针对性。结论:从传统复方中筛选活性单体可能乃中西医结合之新方向。更多还原

【Abstract】 Traditional Chinese medicine(TCM) is characterized by holistic concept and treatment according to syndrome differentiation and the prescriptions of TCM play a good effect in the treatment of disease.As it has multiple components and multiple target points,the modernization of TCM encounters obstacles.Thus it is particularly important for us to find activate molecule from prescriptions.Method:With the rise of systems biology,it becomes more and more popular to screening active molecule based on biological database for foreign researchers in recent years.Inspired by the classical theory of TCM,our group put forward a new method of screening activate molecule from prescriptions,namely a "component-target gene-disease" excavation based on systematic biology.A variety of database are used to screen the activate molecule which is the key in the treatment of disease from prescriptions of TCM,thus we can finish the abutment between "activate molecule" and "target gene" point to point and conduct the experiment to validate the effect of the screened effective molecule.Result:This method can achieve the screening of "Activate molecule" from "Jun-Chen-Zuo-Shi" prescriptions.Compared with the screening methods of foreign researchers,this method can be used to find the active molecule from TCM prescriptions with definite clinical efficacy,which can greatly reduce the scope of screening.Conclusion:It is a new direction of combination of traditional Chinese and Western medicine to screen active molecule from traditional prescriptions.更多还原
