TUNEL法探讨以“肝脾”为核心对高催乳激素血症模型大鼠卵巢颗粒细胞凋亡的影响

【摘要】 目的:运用TUNEL法探讨以"肝脾"为核心对高催乳激素血症(Hyperprolactinemia,HPRL)模型大鼠卵巢颗粒细胞凋亡的影响。方法:将SD成熟雌性大鼠,随机分为作空白对照组、模型组、溴隐亭组、中药高、中、低剂量组。除空白对照组外,其余组用甲氧氯普胺腹腔注射制作HPRL大鼠模型,各组完成相应30天灌胃治疗后,处死开腹取卵巢,运用TUNEL法检测凋亡阳性细胞数。结果:中药高剂量组能显著降低凋亡小体,P<0.01,各剂量组呈量效关系;颗粒细胞生长量多、排列整齐、形态规则;中剂量组次之;低剂量组较差;结论:逍遥散加减方可降低凋亡小体的表达,改善卵巢颗粒细胞的形态,减少颗粒细胞凋亡。
【Abstract】 Objective: Using TUNEL to discuss the effect of Hyperprolactinemia model rat ovarian granulosa cells apoptosis with core of "liver and spleen". Methods: Mature female SD rats, and randomly divided into blank control group, model group, bromocriptine group, high、medium and low dose Chinese medicine groups. In addition to the blank control group, the rest of the group use theintraperitoneal injection of metoclopramideto production HPRL rat model, by 30 days gavage treatment of each group, sacrificed and take the ovary, by using TUNEL method to detect apoptosis positive cells number. Results: High dose group can significantly reduce the apoptotic bodies, P < 0.01, each dose group of the concentration-response relationship; Granulosacells growth, regular arrangement and morphology; Middle dose group took second place; Low dose group was the worst; Conclusion: Xiaoyao powder can reduce the expression of apoptotic bodies, improve ovarian granulosa cell morphology, reduce granulosa cell apoptosis.
