MBI as a forerunner of progression to Alzheimer’s disease

【摘要】 Cognitive impairment, in addition to deposition of beta-amyloid plaques and neurofibrillary tangles, is the clinical hallmark of Alzheimer’s disease(AD). Mild cognitive impairment(MCI) has been associated with risk for Alzheimer’s disease since people with MCI are more likely to develop AD than people without MCI. However, it’s not realized until recently that mild behavioral impairments(MBI) are often first signs of dementia. MBI occurring at very early age of AD patients may involve abnormal emotion, social activity and stereotyped behavior, etc. There is no doubt that MBI may be a forerunner of neurodegeneration and progression to MCI and even AD, but the diagnostic criteria of MBI and underlying neural network and molecular mechanisms remained largely unknown. Utilizing transgenetic AD mouse model and littermate controls, we observed increased anxiety, altered sociability and repetitive behavior in young AD mice. In addition to behavioral changes, AD mice showed impaired prefrontal cortical(PFC) electrical activities accompanying with altered number of parvalbumin(PV) interneurons. Together, the study suggests that alterations in GABAergic signaling pathway may play an important role in the development of MBI.
