Role of mating-type genes in sexual reproduction and pathogenicity of Magnaporthe oryzae

【摘要】 Magnaporthe oryzae is the causal agent of rice blast disease,the most devastating of cultivated rice(Oryza sativa) and a continuing threat to global food security.M.oryzae,as a heterothallic fungus,also produces sexual generation,but needs two parent strains carrying opposite mating-types to complete the mating process.M.oryzae has two mating types of species,MAT1-1 and MAT1-2,The MAT1-1 gene locuscontains three genes,MAT1-1-1,MAT1-1-2 and MAT1-1-3,and the MAT1-2 locus contains two genes,MAT1-2-1 and MAT1-2-2.In the present work,we investigated the roles of the five mating genes,in sexual reproduction and pathogenicity of M.oryzae.Via homologous recombination,MAT1-1-1,MAT1-1-2 and MAT1-1-3 were respectively deleted from a M.oryzae MAT1-1 strain,and MATl-2-1 and MAT1-2-2 were deleted from a M.oryzae MAT1-2 strain.The phenotypical analysis indicated that MAT1-1-1 and MAT1-1-3 were involved in vegetative growth,MAT1-1-1,MAT1-1-2,MAT1-1-3 participated in sporulation,and MAT1-2-2 mutant was likely affected in cell wall integrality.Crossing-cultured with a strain in opposite mating type,△mat1-1-1,△mat1-1-2,△mat1-1-3 and △mat1-2-1,other than △mat1-2-2,formed much fewer perithecia,suggesting that the MAT1-1-1,MAT1-1-2,MAT1-1-3 and MAT1-2-2 are required for sexual reproduction in M.oryzae.However,inoculation test showed that the pathogenicity was unaltered in these mutants.
