母亲肠道细菌多样性及其功能基因研究

【摘要】 怀孕前后母亲肠道微生态发生显著适应性改变,孕期特征显著且可在妊娠结束后得以维持。反应自身健康状况的同时还对新生儿肠道菌群的定殖与发育,以至对成年以后的代谢与免疫疾病的发生产生重要影响。本文采用单细胞扩增技术与宏基因组方法相结合的策略以期全面了解产妇肠道细菌多样性及其功能基因组成。对来自五位健康产妇粪便样本测序发现,共享核心菌门4个,依次为厚壁菌门(53.43%)、拟杆菌门(38.60%)、变形菌门(5.46%)和柔膜菌门(1.59%);共享核心菌属14个,占样品微生物总量的77.99%。基因功能涉及转运因子、ABC转运体、DNA修复和重组蛋白、核糖体、嘌呤代谢、蛋白酶、嘧啶代谢、转录因子、氨基糖和核苷酸糖代谢、未知功能、双组分系统等306个功能大类,功能热图分析显示样本间功能基因分布存在差异。多糖代谢到短链脂肪酸生成相关功能基因分析显示,多菌属可为难消化多糖提供增补遗传代谢途径,其中拟杆菌门和厚壁菌门相关菌属分别在多糖降解和短链脂肪酸生成表现各自优势,菌间合作为营养及能量高效摄取提供可能。本文为进一步理解母亲微生物群落结构与功能提供参考。

【Abstract】 The mcrobiota composition of healthy mother changes adaptively during pregnancy which can last to lactation. It reflects the maternal health condition and the colonization and development of neonatal intestinal flora which can further affects the metabolism and immune disease of adulthood.In our article,single cell amplification technique combined with macrogenomics were used for fully understanding the diversity and functions in the mcrobiota of healthy mother intestinal. The four shared core phylum were firmicutes(53.43%),bacteroidetes(38.60%),proteobacteria(5.46%) and tenericutes(1.59%). The 14 main shared genus accounts for 77.99% of the total intestinal bacteria. 306 functions were selected in the samples,which innvolving transfer factor, ABC transporter, DNA repair and recombinant protein, ribosomes, purine metabolism, protease, pyrimidine metabolism, transcription factors, metabolism of amino and sugars, unknown functions compared with two-component system.Functional heatmap analysis showed differences in the distribution of functional genes between samples.The analysis of functional genes involving in polysaccharide metabolism to short chain fatty acids shows that many species provide a supplementary genetic metabolic pathways for polysaccharides that are difficult to degrade hydrolyze,among them,some genus of bacteroides and firmicutes were related to polysaccharide degradation and short-chain fatty acid production respectively.The crossfeeding between bacteria groups provides the possibility for obtaining energy efficiently. The paper provides a reference for understanding the structure and function of mother microbial community.
