双歧杆菌中10-羟基-顺-12-十八碳烯酸通过可逆生成亚油酸方式转化合成共轭亚油酸

【摘要】 10-羟基-顺-12十八碳烯酸(10-HOE)是亚油酸(LA)10号碳上含有羟基的单烯酸。乳杆菌中的油酸水合酶可以催化LA生成10-HOE,作为共轭亚油酸(CLA)合成的的第一个中间产物;双歧杆菌来源的油酸水合酶也可以催化LA生成10-HOE,但却被证明不参与CLA的合成。目前10-HOE在双歧杆菌生物合成CLA过程中的发挥的作用至今未得到有效阐述。本文讨论双歧杆菌生物合成CLA过程中,LA,CLA以及10-HOE三个物质的动态变化。结果显示双歧杆菌可以利用LA以及10-HOE作为底物转化生成CLA,但是LA可以直接作为底物转化生成CLA以及10-HOE,但是CLA的积累早于10-HOE的合成;但是10-HOE必须首先转化成LA,然后才能进一步被转化生成CLA。油酸水合酶和亚油酸异构酶不仅在利用LA方面存在竞争关系,前者还可通过催化10-HOE产生LA,进一步影响CLA的合成。本文是首次报道双歧杆菌中10-HOE参与CLA合成的方式。

【Abstract】 10-hydroxy-cis-12 octadecaenoic acid(10-HOE) is a kind of octadecaenoic acid with hydroxyl on the C10 carbon. Oleate hydratase derived from Lactobacil i could transform LA into 10-HOE, as the first intermediate during CLA biosynthesis, while this enzyme from Bifidobacterium was identified to be not responsible for CLA production, though it could could also catalyze 10-HOE production. The precise function of 10-HOE during this process is still unknown. Dynamic changes of LA, CLA and 10-HOE during CLA-producing process by Bifidobacterium was discussed. Results demonstrated that both of LA and 10-HOE could be used as the substrate by these strains to transform into CLA but via different pathway. LA could be transferred into CLA directly, while 10-HOE must be firstly catalyzed into LA by oleate hydratase and then isomerized into CLA. Competition of LA existed between oleate hydratase and linoleic acid isomerase, but the former could also influence the CLA accumulation by transforming 10-HOE into LA. This is the first report of the CLA-producing way that 10-HOE is involved in among Bifidobacterium.
