活性小分子细胞、活体成像研究进展

【摘要】 生物活性小分子调控细胞功能,并与重大疾病的发生发展密切相关。由于活性小分子往往浓度低、寿命短、存在相互关联转化,并且在细胞及活体的不同部位出现。因此,发展高灵敏、动态、原位、多组分准确检测活性小分子的技术,对深入剖析重大疾病的发生发展的分子机制,实现早期诊治至关重要。荧光成像技术具有高灵敏度、高时空分辨、低生物毒性的优点,是精准检测细胞及活体内活性小分子的有力工具,但目前该技术所需要的理想的荧光探针仍然缺乏。为解决上述问题,我们分别发展了高灵敏度荧光探针、动态可逆荧光探针、多组分检测荧光探针及亚细胞区域定位荧光探针,对细胞及活体内的生物活性小分子进行了荧光成像研究。以上工作取得的结果,为彻底了解肿瘤等重大疾病发生发展的分子机制,新药的开发与疾病诊治研究提供了有力的技术支撑。

【Abstract】 Biologically active small molecules play vital roles in numerous cellular processes. Emerging studies suggest that they are closely linked with occurrence, development of diseases. Therefore, accurate detection of them can greatly promote discovery of molecular mechanism of relevant diseases. Considering its unique characters including extremely low concentration, very short life, generation in different cellular compartments and correlation with other biological molecules, based on specific recognition reactions, we constructed a series of new fluorescent probes that possess ultra-high sensitivity, reversibility, organelle-targeting and multicomponent-labeling ability, and also developed new principles and methods for real-time and in situ fluorescence imaging of biologically active small molecules. The obtained results give new insight into its regulation roles of cell functions, which contribute to the study of early diagnosis and therapy of related major diseases.
