具有聚集诱导发光性质的荧光增强型鱼精蛋白探针

【摘要】 利用Diels-alder反应,我们用简单廉价的原材料设计合成了荧光增强型检测鱼精蛋白探针:TPAA。由于分子内旋转受限(RIR)机理,TPAA在水/THF混合体系中荧光强度随水含量的增加而增强,表现出明显的聚集诱导发光(AIE)性质。同时,在不同pH条件下,由于质子化与去质子化作用,TPAA在不同p H中的聚集方式发生变化导致其荧光强度随pH的增大而降低。我们研究了在高p H时TPAA对鱼精蛋白的检测行为。在pH=9时,随着鱼精蛋白的加入,TPAA的荧光强度逐渐增强,在加入约6μg/mL时,荧光强度几乎达到最大值,此时的荧光强度是原始空白样品荧光强度的45倍。检测限为4.78 ng/mL。选择性实验说明,不论其他蛋白存在与否,对检测鱼精蛋白均不会产生影响。由于肝素与鱼精蛋白的结合作用大于TPAA与鱼精蛋白的结合,因此该体系还可用于检测肝素的含量。
【Abstract】 Fluorescence turn-on protamine probe,namely TPAA,was designed and synthesized using costless raw materials according to Diels-alder action.The fluorescence intensity of TPAA increased as water content increased because of restricted intramolecular rotation(RIR).At the same time,the fluorescence intensity of TPAA decreased as pH values increased,because the aggregation behavior of TPAA depends on the pH conditions due to the protonation-deprotonation process.The binding behavior between TPAA and protamine was studied.The fluorescent intensity increased as the amount of protamine increased.The FL intensity is 45 times higher than the original FL intensity when 6 μg/mL was added.The detection limit is 4.78 ng/mL.The detection behavior was not effected whether other proteins in the present of the detection system or absent.Heparin is the most negatively charged biological macromolecule and can strongly bind with protamine through electrostatic interaction,thus,TPAA could also be utilized to investigate the interactions between protamine and heparin.
