【摘要】 运用石蜡切片H&E染色技术和PCR技术,调查分析牛和羊心肌(黄牛405份,绵羊311份)食源性住肉孢子虫病的流行情况和感染强度。结果发现,黄牛住肉孢子虫感染率为49.38%(200/405,95%CI=44.54-54.23),感染密度为5.17~8.73个/cm2,绵羊住肉孢子虫的感染率为32.48%(101/311,95%CI=27.51-37.87),感染密度为7.06~32.02个/cm2。其中黄牛住肉孢子虫薄壁包囊占88.00%,厚壁包囊2.00%,混合感染10.00%,而绵羊住肉孢子虫薄壁包囊、厚壁包囊、混合感染的感染率分别为38.61%,31.68%,29.71%。牛、羊心肌消化液成功扩增出DNA长度为350 bp的牛住肉孢子虫基因片段、羊住肉孢子虫600 bp的基因片段,进而验证形态学结果。病理组织学观察和PCR结果显示,牛和羊心肌中分别存在住肉孢子虫S.cruzi、S.hominis和S.arieticanis、S.tenella,均对牛羊存在致病性,并且危害肉品质量安全。该研究为防控牛羊住肉孢子虫病,牛、羊肉品的安全监测提供参考依据。中原地区牛、羊心肌住肉孢子虫病流行情况首次报道。更多还原

【Abstract】 In order to investigate the epidemiological of sarcocystosis in cattle and sheep.The infection intensity of sarcocystis in myocardium of cattle(405) and sheep(311) was analyzed by H&E staining and PCR in this study.The infection rate was 49.38%(200/405,95%CI=44.54-54.23) in cattle,infection density was 5.17~8.73 per/cm2.The infection rate was32.48%(101/311,95% CI=27.51-37.87),infection density was 7.06~32.02 per/cm2 in sheep.The percent of thin-walled cyst was 88.00%,thick-walled cyst was 2.00%,and mixed infections was10.00% in cattle.It was 38.61%,31.68%,29.71% respectively in sheep.DNA from myocardial digestive succeed amplified 350 bp product from cattle,600 bp product from sheep,and then verified the morphological results.Histopathological and PCR results showed that S.cruzi and S.hominis,S.arieticanis and S.tenella were present in cattle and sheep myocardium.They had pathogenicity on cattle and sheep,and posed threaten on the quality and safety of meat.The study provided reference basis for the prevention and control of cattle and sheep sarcocystosis,and in meat safety monitoring.This is the first research of sarcocystiosis from cattle and sheep in the central region of China.更多还原
