基于线粒体COI基因的DNA条形码在南极鱼科和鳄冰鱼科鱼类分类中的应用

【摘要】 本文采用CO1通用引物扩增测序并结合GenB ank已有序列联配分析,对南极鱼科和鳄冰鱼科2科22属43种共98条CO1基因片段(539bp)进行序列比较和系统进化研究。结果分析表明43种南极鱼科和鳄冰鱼科的鱼类COI基因的平均G+C含量为45.9%,具有明显的碱基偏倚性。南极鱼亚目两个科鱼类的种间平均距离(0.157)是种内平均距离(0.002)的79倍,南极小带腭鱼(Cryodraco antarcticus)和罗斯海小带腭鱼(Cryodraco atkinsoni)的种间遗传距离为0.002,推断二者为同种异名。从分子系统树看出,39个种(90.70%)可以独立成支且与形态分类结果一致。由此可见,DNA条形码不仅能够对遗传距离较近(0.021-0.028)的物种可准确区分,并且对南极鱼亚目鳄冰鱼科和南极鱼科鱼类能够进行有效地物种鉴定。总之,DNA条形码不仅可以作为形态学的辅助手段,为南极鱼分类系统的必要补充和佐证并且可以用于探讨南极鱼类的系统发育关系。
【Abstract】 The order Notothenioidei and Channichthyidae in the Class Osteichthyes, and many of these species are of significant economic value.The mitochondrial cytochrome c oxidase subunit 1(COI) gene sequences of fifteen species pertaining to six genera in Notothenioidei and Channichthyidae collected from the Southern Ocean were amplified using a polymerase chain reaction technique. In this study the homologous sequences of other species of Notothenioidei and Channichthyidae from Gen Bank. A total of 98 single sequences from 43 species pertaining to 22 genera in Notothenioidei and Channichthyidae were barcoded using the COI gene and then sequenced and compared to verify the feasibility of using DNA barcoding to identify species.Our results showed that the COI sequence of 43 species of Antarctic fishes were obviously base biased( average G+C 45.9%). In Notothenioidei, average genetic distance(0.157) among species is 79 times as big as within species(0.002),Cryodraco antarcticus and Cryodraco atkinsoni genetic distance(0.002) suggesting that these may be the same species. From the phylogenetic tree, the 39 species can cluster respectively, which were consisted with the morphological classification. Thus, we can conclude that COI DNA barcode can effectively identify the remaining 39 species.Generally, COI barcode can not only as an assisted mean of species identification but also can used for phylogenetic classification in Antarctic fish.
